Ab initio HF, density functional and experimental studies on the IR spectra and structure of 1,2-benzisothiazol-3-(2H)-thione-1,1-dioxide (thiosacchanin) and its nitranion

The spectral and structural changes taking place in the course of the conversion of 1,2-benzisothiazol-3-(2H)-thione-1,1-dioxide (thiosaccharin) into a nitranion have been studied on the basis of both IR spectra and ab initio HF 6-31G(d) and BLYP 6-31G(d,p) force field calculations. The conversion causes nuSO(2)as and nuSO(2)s frequency decreases of 47 and 13 cm-1, respectively, and other spectral changes. The nuC double bond S coordinate is strongly delocalized. The ab initio geometries of the isolated molecule and nitranion agree well with the single-crystal X-ray ones, determined for thiosaccharin and its sodium (potassium) monohydrate salts, respectively. The nitranionic charge is delocalized almost uniformly within the thiocarbonyl (0.29 e-), sulfonyl (0.24 e-), and phenylene (0.24 e-) groups, and the nitranionic center (0.23 e-).

Analysis of volatiles in brown rice, germinated brown rice, and selenised germinated brown rice during storage at different vacuum levels

BACKGROUND: The quality of nutritionally enhanced foods can be determined by evaluating changes in the volatile compounds produced in these foods over time. In this work, selenium-enriched germinated brown rice (Se-GBR), germinated brown rice (GBR), and brown rice (BR) stored under 90% relative humidity, 38 C, and various vacuum levels were investigated. The relative abundance and differences of volatile compounds in Se-GBR, GBR, and BR over various storage periods were detected. The correlation of volatile compound abundance with vacuum level and storage time was analysed using principal component analysis (PCA). RESULTS: Volatile compounds in the three samples were quantified at various storage periods (0, 90 and 150 days). Approximately 100 volatile compounds and eight species were identified and classified. Various proportions or types of volatile compounds were found in each sample at different sampling times. PCA results showed an isolation of volatile compounds in terms of sampling day and vacuum level at each storage period. CONCLUSION: Changes in volatile compounds over time and vacuum levels can provide bases for assessing of the nutritional quality of Se-GBR, GBR, and BR.

Aldehyde oxidase (AO) is a cytosolic enzyme that belongs to the family of structurally related molybdoflavoproteins like xanthine oxidase (XO). It catalyzes the oxidation of aromatic and aliphatic aldehydes and various heteroaromatic rings as well as reduction of several functional groups. Several reviews on the role of AO in drug metabolism have been published in the past decade indicative of the growing interest in the enzyme and its influence in drug metabolism. Here, we present a comprehensive monograph of AO as a drug metabolizing enzyme with emphasis on marketed drugs as well as other xenobiotics, as substrates and inhibitors. Although the number of drugs that are primarily metabolized by AO are few, the impact of AO on drug development has been extensive. We also discuss the effect of AO on the systemic exposure and clearance these clinical candidates. The review provides a comprehensive analysis of drug discovery compounds involving AO with the focus on developmental candidates that were reported in the past five years with regards to pharmacokinetics and toxicity. While there is only one known report of AO-mediated clinically relevant drug-drug interaction (DDI), a detailed description of inhibitors and inducers of AO known to date has been presented here and the potential risks associated with DDI. The increasing recognition of the importance of AO has led to significant progress in predicting the site of AO-mediated metabolism using computational methods. Additionally, marked species difference in expression of AO makes it is difficult to predict human clearance with high confidence. The progress made towards developing in vivo, in vitro and in silico approaches for predicting AO metabolism and estimating human clearance of compounds that are metabolized by AO have also been discussed.

Members of the series of title compounds were tested for potential antipsychotic activity in relevant receptor binding assays and behavioral screens. Structure-activity relationships within the series are discussed. Compound 24 (BMY 13859-1), a (1,2-benzisothiazol-3-yl)piperazine derivative, was selected for further study because of its potent and selective profile in primary CNS tests. It was active in the Sidman avoidance paradigm and blocked amphetamine-induced stereotyped behavior in dogs for up to 7 h. The compound’s lack of typical neuroleptic-like effects in the rat catalepsy test and its failure to produce dopamine receptor supersensitivity following chronic administration indicate that it should not cause the movement disorders commonly associated with antipsychotic therapy. Although 24 has potent affinity for dopaminergic binding sites, its even greater affinity for serotonin receptors suggests that a serotonergic component may be relevant to its atypical profile. Compound 24 is currently undergoing clinical evaluation in schizophrenic patients.
