Cyano substituent effects on enol and enethiol acidity and basicity: The protonation and deprotonation of 3-hydroxy-2-propenenitrile and its thio analogue

The gas-phase basicity and acidity of 3-hydroxy-2-propenenitrile (3-hydroxyacrylonitrile) and its sulfur-containing analogue, 3-mercapto-2-propenenitrile, have been determined by means of high-level G3B3 ab initio calculations and, in the case of the latter compound, compared with the experimental values obtained by means of FT-ICR mass spectrometry techniques, and with previous reported values for the N{triple bond, long}C-CH{double bond, long}CH-X (X = CH3, NH2, SiH3, PH2) analogues. For both compounds the Z-isomer is the dominant species in the gas-phase. Protonation takes place in both cases at the cyano group. The loss of the proton from the substituent, was found to be systematically much more favorable than the deprotonation at the HC{double bond, long}CH group. 3-Hydroxy-2-propenenitrile is predicted to be a stronger base by ca. 5 kJ mol-1 than its thio analogue, but a weaker acid by 26 kJ mol-1. Both compounds are stronger acids than the corresponding unsubstituted vinyl compounds, because cyano substitution stabilizes much more the deprotonated species than the corresponding neutral compound. There is a clear disagreement between our theoretical estimates for both the gas-phase basicity and the gas-phase acidity of 3-mercapto-2-propenenitrile and the corresponding experimental values, which is consistent with its isomerization to yield isothiazole.

Aroma characterization of red wines from cv. Bobal grape variety grown in La Mancha region

Bobal is an indigenous Spanish cultivar, principally grown in the region of Valencia. The volatile composition of young red wines made from the cv. Bobal grape variety grown in the La Mancha region of Spain has been studied over five vintages by instrumental analysis to determine the influence of grape variety on the aroma of wine. Free and glycosidically-bound aroma compounds were isolated by solid phase extraction (SPE) using dichloromethane and ethyl acetate respectively to later be analyzed using gas chromatography-mass spectrometry (GC/MS). One hundred and twelve (112) free aroma compounds and ninety-six (96) bound aroma compounds were identified and quantified in Bobal wines oven this five year period. Based on the result, Bobal wine presents a complex chemical profile with a wealth of aromas in its aromatic composition.

The crystal structure of pyridinium thiosaccharinate thiosaccharin, [HNC5H5]+ [(tsac)(Htsac)]- (Htsac = C7H5NO2S2, the thiosaccharin molecule), was determined by single-crystal X-ray diffraction. Both, the thiosaccharinate anion, the corresponding neutral molecule and the pyridinium cation are nearly planar and arranged in the solid state in an almost parallel fashion, hence giving rise to a layered supramolecular structure. The anion shows small but significant modifications in the bonding of the thioamide group as compared with the neutral molecule. The crystal is further stabilized by an extensive H-bonding network. The FTIR spectrum of the compound is briefly commented.

Volatile and sensory characterization of red wines from cv. Moravia Agria minority grape variety cultivated in La Mancha region over five consecutive vintages

In this work, the wines were elaborated across five harvests (2004-2008) from Moravia Agria minority red grape variety cultivated in La Mancha region. Moravia Agria wines were studied by instrumental and sensory analysis to determine the influence of grape variety on the aroma of wine. Free and glycosidically bound aroma compounds were isolated by solid phase extraction (SPE) to later be analyzed using gas chromatography-mass spectrometry (GC/MS). Ninety-two (92) free aroma compounds and sixty-seven (67) bound aroma compounds were identified and quantified in Moravia Agria wines over this five year period. The odor activity values (OAVs) for the different compounds were classified into seven odorants. The fruity and sweet series are the ones most strongly contributing to the aroma profile of Moravia Agria wines. The sensory profile of Moravia Agria wines was evaluated by experienced wine-testers and was characterized by red fruit, fresh, citric, flower and lychee aroma descriptors. This study shows the first complete aromatic characterization of Moravia Agria wines, also the data suggest that these wines present a great aromatic potential.

The chemistry of heterocyclic compounds plays a crucial role in the synthesis of medicinals. This review focuses on the use of the thiazole nucleus for the synthesis of newer drug molecules through rational drug discovery. Here the synthetic feasibility, biochemical compatibility and the therapeutic utility of the thiazole derivatives is discussed briefly. Recently, it was observed that many chemotherapeutic agents have a thiazole nucleus. Hence, this article highlights the profound anti-cancer activities of some major thiazole bearing drug molecules with their important target sites. Along with this, the recent advancements in the development of thiazole based newer anti-cancer molecules and their promising activities are reviewed. The relevant data and some statistical analysis regarding the medicinal importance of thiazole nucleus will further promote the design and development of varieties of chemotherapeutic entities in the field of cancer treatment.

Novel urea and thiourea derivatives of thiazole-glutamic acid conjugate as potential inhibitors of microbes and fungi

Since discovery and development of effective as well as safe drugs has brought a progressive era in human healthcare that is accompanied by the appearance of drug resistant bacterial strains, there is constant need of new antibacterial agent having novel mechanisms of action to act against the harmful pathogens. In the present study, several N-terminal substituted urea/thiourea derivatives were synthesized by the reaction of glutamic acid and 3-(1-piperazinyl)-1,2-benzisothiazole with various substituted phenyl isocyanates/isothiocyanates. Elemental analysis, IR, 1H NMR, 13C NMR and mass spectral data confirmed the structure of the newly synthesized compounds. The derivatives were investigated for their antibacterial and antifungal activities against various pathogens of human origin by agar well diffusion method and microdilution method. The preliminary antimicrobial bioassay reveals that the compounds containing fluoro and bromo as substituents showed promising antimicrobial activity. Pleiades Publishing, Ltd., 2013.
