Quantification of Aromaticity of Heterocyclic Systems Using Interaction Coordinates

Recently, we proposed an aromaticity index based on interaction coordinates (AIBIC) (J. Phys. Chem. A 2016, 120, 2894a’2901). This index works well for the aromatic hydrocarbons. However, in the case of heterocyclic systems, the AIBIC overestimates the aromaticity indicating many of them to be more aromatic than benzene, which seems unlikely. Because of the differences in the electronegativity of the carbon and the other heteroatoms, the electron density is partially localized near the more electronegative atom(s) of the aromatic fragment. This localized electron density does not contribute to the aromaticity that is due to the delocalized electron density over the central ring. To account for this reduction in the delocalized electron density, a correction is introduced based on Pauling’s electronegativity equation. When the corrected interaction coordinates are used in the computation of AIBIC, we get a new index-aromaticity index based on interaction coordinates corrected. This new index, when computed for a variety of heterocyclic systems, yields results in line with the expectations, and its usefulness in quantifying aromaticity appears to be very promising.

Concentrations, sulfur isotopic compositions and origin of organosulfur compounds in pore waters of a highly polluted raised peatland

Sulfur cycling in peatlands plays a key role in their response to atmospheric deposition of acid sulfate; however, the role of dissolved organic sulfur species has received little attention. In this study, we assess the quantitative importance of dissolved organic sulfur species in a heavily polluted peatland and their role in sulfur cycling. Surface and pore waters from a raised peat bog have been analyzed for dissolved organic and inorganic species. Except in the surface water, organically bound sulfur dominates over sulfate sulfur and the organosulfur is predominantly in the humic fraction, which is not GC amenable. Two organosulfur compounds are present in the GC amenable fraction at all depths; a compound containing a benzene ring, sulfone and amide groups dominates while a compound closely similar to either benzothiazole or 1,2-benzisothiazole is present at 100-1000 times lower concentrations. Other organosulfur species were detected in some samples by GC-AED (gas chromatography-atomic emission detection) at similar or yet lower concentrations but not identifiable with GC-MS. In the shallowest parts of the peat, sulfur isotopic data on the humic and non-humic dissolved organic sulfur fractions show a pronounced difference, implying different sources for these organosulfur compounds. Humic organosulfur is 34S enriched, indicating a source for this component from breakdown of plant materials, which have similar delta34S (0.1? and 4.2?). By contrast, the non-humic organosulfur is more 34S depleted at shallow depths (-11.7? in surface water), indicating an origin from reaction of bacterially produced sulfide (-9.9? in surface water) with dissolved organic material. Deeper in the peat the non-humic organosulfur becomes more 34S enriched (up to +6.8?), which may result from either: (i) reactions between organic material and more 34S enriched sulfide; or (less likely) (ii) a shift to production of non-humic organosulfur by breakdown of humic compounds or plant material in the peat matrix. These data highlight the importance of organosulfur species relative to inorganic species in peatland groundwaters. Organosulfur species comprise the bulk of the dissolved sulfur budget and are involved in the bacterial cycling of sulfur in the peatland ecosystem.

Toxicity and mutagenicity of low-metallic automotive brake pad materials

Organic friction materials are standardly used in brakes of small planes, railroad vehicles, trucks and passenger cars. The growing transportation sector requires a better understanding of the negative impact related to the release of potentially hazardous materials into the environment. This includes brakes which can release enormous quantities of wear particulates. This paper addresses in vitro detection of toxic and mutagenic potency of one model and two commercially available low-metallic automotive brake pads used in passenger cars sold in the EU market. The model pad made in the laboratory was also subjected to a standardized brake dynamometer test and the generated non-airborne wear particles were also investigated. Qualitative “organic composition” was determined by GC/MS screening of dichloromethane extracts. Acute toxicity and mutagenicity of four investigated sample types were assessed in vitro by bioluminescence assay using marine bacteria Vibrio fischeri and by two bacterial bioassays i) Ames test on Salmonella typhimurium His- and ii) SOS Chromotest using Escherichia coli PQ37 strain. Screening of organic composition revealed a high variety of organic compounds present in the initial brake pads and also in the generated non-airborne wear debris. Several detected compounds are classified by IARC as possibly carcinogenic to humans, e. g. benzene derivatives. Acute toxicity bioassay revealed a response of bacterial cells after exposure to all samples used. Phenolic resin and wear debris were found to be acutely toxic; however in term of mutagenicity the response was negative. All non-friction exposed brake pad samples (a model pad and two commercial pad samples) were mutagenic with metabolic activation in vitro.

Aerogel from chemo-enzymatically oxidized fenugreek gum: an innovative delivery system of isothiazolinones biocides

Aerogels of chemo-enzymatically oxidized, lyophilized fenugreek gum (EOLFG) were evaluated as ?delivery system? (DS) of the microbiocide mix 5-chloro-2-methyl-4-isothiazolin-3-one (CIT) and 2-methyl-4-isothiazolin-3-one (MIT). These biocides have a broad activity spectrum and a low MIC (minimal inhibitory concentration) in the ppm range. They are approved under the EU Biocidal Product Directive for various applications, including cosmetics, are classified as skin sensitizers, and are under increasing scrutiny to limit or eliminate their use. We demonstrate that CIT/MIT can be uptaken into EOLFG aerogel corks by immersion into aqueous solutions of microbiocides, followed by blotting and re-lyophilization to generate ?loaded? EOLFG aerogels. Incubation of ?loaded? corks in water brings about a slow and almost complete release of the absorbed actives having the same MIC of free biocides against selected bacterial pathogens. This new biomaterial could represent an innovative DS of microbiocides and other actives for a variety of industrial, food, cosmetic, and biomedical applications.

Organic compounds containing five-membered aromatic heterocyclic rings play an important role in life science. Among them thiazole and isothiazole heterocycles are present in many pharmaceutical drugs and crop protection compounds and have sparked over past years an enormous interest on these scaffolds. In agrochemicals these moieties are found in 11 commercial products as well as in two development compounds. A thiazole moiety is present in seven currently marketed agrochemicals, namely the insecticides thiamethoxam and clothianidin, the fungicides thiabendazole, thifluzamide, ethaboxam, and oxathiapiprolin, and the nematicide fluensulfone, as well as in the development compound dicloromezotiaz. One compound, the fungicide isotianil, contains an isothiazole ring and three compounds a benzothiazole ring, namely the fungicide benthiavalicarb and the herbicides methabenzthiazuron and mefenacet. A benzisothiazole ring is not yet present in any commercial product, however, is found in the former nematicide development compound benclothiaz. Interestingly thiazole and isothiazole heterocycles are much less frequently found in agrochemicals than pyridine and pyrazole despite their bioisosteric relationship, which may indicate that these heterocycles have still not been explored to their full scope in crop protection lead discovery and optimization. Detailed information on the discovery, synthesis, and the structure?activity relationships of thiazole- and isothiazole-containing agrochemicals is provided in this review.
