A novel path has been developed for the conversion of carboxylic acids into the corresponding N -acylbenzotriazoles by using 2,2-dipyridyl disulfide/PPh (3) in anhydrous dichloromethane in the presence of 1 H -benzotriazole. Mild reaction conditions, short reaction time, easy in handling, wide substrate scope, availability of reagents involved, and moreover avoiding the use of base makes this protocol quite useful for the laboratory practices for N -acylbenzotriazole synthesis.

alpha-Aryl oxindoles are accessed from isatin via a two-step procedure involving a phospha-Brook rearrangement and a Friedel-Crafts alkylation in a one-pot procedure. The use of 1,1,1,3,3,3-hexafluoro-2-propanol as solvent significantly extended the reaction substrate scope to include relatively less electron-rich arenes including benzene. This new alkylation method is fast and straightforward and allows for the direct introduction of the oxindole moiety onto a range of aromatic compounds including phenols. Additionally, the application of arylated products was shown in decarboxylative asymmetric allylation and protonation.

We describe the preparation of two monomers that bear complementary nucleobases at the edges (guanine-2′-deoxycytidine and 2-aminoadenine-2′-deoxyuridine) and that are conveniently protected and activated for solid-phase automated DNA synthesis. We present an atom-economic strategy to catalytically generate and intercept nitrile anion equivalents using hydrogen transfer catalysis. Addition of alpha,beta-unsaturated nitriles to a pincer-based Ru-H complex affords structurally characterized kappa-N-coordinated keteniminates by selective 1,4-hydride transfer. When generated insitu under catalytic hydrogenation conditions, electrophilic addition to the keteniminate was achieved using anhydrides to provide alpha-cyanoacetates in high yields. This work represents a new application of hydrogen transfer catalysis using alpha,beta-unsaturated nitriles for reductive C-C coupling reactions.

Herein, we report a redox-neutral Ni-catalyzed cross-coupling reaction of two readily available precursors to produce the corresponding ketones that are an important structural motif in numerous biologically active entities. By the use of a commercially available nickel/Triphos catalytic system, a range of easily accessible alkenyl primary alcohols and aryltriflates can be converted in a rapidly assembled fashion to valuable ketones with good yields and wide functional group tolerance. We also demonstrate the utility of this transformation by late-stage functionalization of a large set of complex molecules with good efficiency, which offers a distinct entry to more functionalized aromatic ketones.

The chlorination of a bioactive compound can change its physiological properties and improve its pharmacokinetic and pharmacological profiles. It therefore has been an important strategy for drug discovery and development. However, the direct aromatic chlorination of complex bioactive molecules is too difficult to be practical. In fact, many functional groups such as hydroxyls, amines, amides or carboxylic acids may strongly restrain the reactivity of Cl+ by forming a halogen bond. Here we report a highly efficient aromatic chlorination of arenes that is catalysed by dimethyl sulfoxide with N-chlorosuccinimide as the chloro source. The mild conditions, easy-availability and stability of the catalyst and reagents, as well as good functional-group tolerance, showed the approach to be a versatile protocol for the late-stage aromatic chlorination of complex natural products, drugs and peptides. The multi-gram experiment and low-cost of N-chlorosuccinimide and dimethyl sulfoxide shows great potential for drug discovery and development in industrial applications. Late-stage aromatic chlorination of active pharmaceutical ingredients has enormous potential in drug discovery yet still features limited applicability due to issues of functional-group tolerance. Now, dimethyl sulfoxide is reported as catalyst for the chlorination of a diverse family of bioactive molecules in combination with N-chlorosuccinimide.

Herein, we have introduced sulfonimidoyl azide as a precursor in a Cu-catalyzed Chan-Evans-Lam reaction with aryl boronic acid for the synthesis of N-aryl sulfonimidamide. The CuCl catalyzed reactions of sulfonimidoyl azides and aryl boronic acids have been proceeded in the presence of triethylamine (Et3N) as base in methanol at room temperature under oxygen atmosphere and resulted moderate to good yields of corresponding products in very short time. Because 6-amino-6-deoxy amylose is an important intermediate for many materials, developing a practical pathway to easily prepare it in bulk is of significance. The synthesis of 6-amino-6-deoxy amylose included three reactions. In the chlorination reaction, the crude product could be fully de-esterified under the improved conditions, and the reaction mechanism was re-understood. The azido group was reduced with sodium borohydride in which the reaction conditions were studied in detail. With the improved synthetic pathway, 6-amino-6-deoxy amylose could be easily prepared. The 6-amino-6-deoxy amylose was characterized via its benzamide derivative with(1)H NMR,H-1-C-13 HSQC NMR, and(1)H-H-1 COSY NMR, confirming the hydroxyl group at C(6)of amylose was converted to amino group with a degree of substitution of 98.2%. Additionally, the 6-amino-6-deoxy amylose was analyzed toward iodine reagent and X-ray diffraction, and the corresponding results showed that it was different from amylose in suprastructure and crystal structure.
