The transition-metal-catalyzed alcohol dehydrogenation to carboxylic acids has been identified as an atom-economical and attractive process. Among various catalytic systems, Ru-based systems have been the most accessed and investigated ones. With our growing interest in the discovery of new Ru catalysts comprising N-heterocyclic carbene (NHC) ligands for the dehydrogenative reactions of alcohols, we designed and prepared five NHC/Ru complexes ([Ru]-1-[Ru]-5) bearing different ancillary NHC ligands. Moreover, the effects of ancillary and additional ligands on the alcohol dehydrogenation with KOH were thoroughly explored, followed by the screening of other parameters. Accordingly, a highly active catalytic system, which is composed of [Ru]-5 combined with an additional NHC precursor L5, was discovered, affording a variety of acid products in a highly efficient manner. Gratifyingly, an extremely low Ru loading (125 ppm) and the maximum TOF value until now (4800) were obtained.

The Mizoroki-Heck reaction is one of the most efficient methods for alkenylation of aryl, vinyl, and alkyl halides. Given its innate nature, this protocol requires the employment of compounds possessing a halogen atom at the site of functionalization. However, the accessibility of organic molecules possessing a halogen atom at a particular site in aliphatic systems is extremely limited. Thus, a protocol that allows a Heck reaction to occur at a specific nonfunctionalized C(sp(3))-H site is desirable. Reported here is a radical relay Heck reaction which allows selective remote alkenylation of aliphatic alcohols at unactivated beta-, gamma-, and delta-C(sp(3))-H sites. The use of an easily installed/removed Si-based auxiliary enables selective I-atom/radical translocation events at remote C-H sites followed by the Heck reaction. Notably, the reaction proceeds smoothly under mild visible-light-mediated conditions at room temperature, producing highly modifiable and valuable alkenol products from readily available alcohols feedstocks.

The utilizations of omnipresent, thermodynamically stable amides and aliphatic C(sp(3))-H bonds for various functionalizations are ongoing challenges in catalysis. In particular, the direct coupling between the two functional groups has not been realized. Here, we report the synergistic activation of the two challenging bonds, the amide C-N and unactivated aliphatic C(sp(3))-H, via metallaphotoredox catalysis to directly acylate aliphatic C-H bonds utilizing amides as stable and readily accessible acyl surrogates. N-acylsuccinimides served as efficient acyl reagents for the streamlined synthesis of synthetically useful ketones from simple C(sp(3))-H substrates. Detailed mechanistic investigations using both computational and experimental mechanistic studies were performed to construct a detailed and complete catalytic cycle. The origin of the superior reactivity of the N-acylsuccinimides over other more reactive acyl sources such as acyl chlorides was found to be an uncommon reaction pathway which commences with C-H activation prior to oxidative addition of the acyl substrate.

To develop antitumor drugs capable of targeting energy metabolism in the tumor microenvironment, we produced a series of potent new biguanide derivatives via structural modification of the arylbiguanide scaffold. We then conducted biological screening using hypoxia inducible factor (HIF)-1- and unfolded protein response (UPR)-dependent reporter assays and selective cytotoxicity assay under low glucose conditions. Homologation studies of aryl-(CH2)(n)-biguanides (n=0-6) yielded highly potent derivatives with an appropriate alkylene linker length (n=5, 6). The o-chlorophenyl derivative 7l (n=5) indicated the most potent inhibitory effects on HIF-1- and UPR-mediated transcriptional activation (IC50; 1.0 +/- 0.1 mu M, 7.5 +/- 0.1 mu M, respectively) and exhibited selective cytotoxicity toward HT29 cells under low glucose condition (IC50; 1.9 +/- 0.1 mu M). Additionally, the protein expression of HIF-1 alpha induced by hypoxia and of GRP78 and GRP94 induced by glucose starvation was markedly suppressed by the biguanides, thereby inhibiting angiogenesis. Metabolic flux and fluorescence-activated cell sorting analyses of tumor cells revealed that the biguanides strongly inhibited oxidative phosphorylation and activated compensative glycolysis in the presence of glucose, whereas both were strongly suppressed in the absence of glucose, resulting in cellular energy depletion and apoptosis. These findings suggest that the pleiotropic effects of these biguanides may contribute to more selective and effective killing of cancer cells due to the suppression of various stress adaptation systems in the tumor microenvironment.

Flavin-based catalysts are photoactive in the visible range which makes them useful in biology and chemistry. Herein, we present electrospray-ionization mass-spectrometry detection of short-lived intermediates in photooxidation of toluene catalysed by flavinium ions (Fl(+)). Previous studies have shown that photoexcited flavins react with aromates by proton-coupled electron transfer (PCET) on the microsecond time scale. For Fl(+), PCET leads to FlH(.+) with the H-atom bound to the N5 position. We show that the reaction continues by coupling between FlH(.+) and hydroperoxy or benzylperoxy radicals at the C4a position of FlH(.+). These results demonstrate that the N5-blocking effect reported for alkylated flavins is also active after PCET in these photocatalytic reactions. Structures of all intermediates were fully characterised by isotopic labelling and by photodissociation spectroscopy. These tools provide a new way to study reaction intermediates in the sub-second time range.

This report details a decarboxylative cross-coupling of (hetero)aryl carboxylates with iodoarenes in the presence of a gold catalyst (>25 examples, up to 96% yield). This reaction is site specific, which overcomes prior limitations associated with gold catalyzed oxidative coupling reactions. The reactivity of the (hetero)aryl carboxylate correlates qualitatively to the field effect parameter (F-ortho). Reactions with isolated gold complexes and DFT calculations support a mechanism proceeding through oxidative addition at a gold(I) cation with decarboxylation being viable at either a gold(I) or a silver(I) species.

Currently, due to the HIV latency mechanism, the search continues for effective drugs to combat this issue and provide a cure for AIDS. Gnidimacrin activates latent HIV-1 replication and inhibits HIV-1 infection at picomolar concentrations. This natural diterpene was able to markedly reduce the latent HIV-1 DNA level and the frequency of latently infected cells. Therefore, gnidimacrin is an excellent lead compound, and its anti-HIV potential merits further investigation. Twenty-nine modified gnidimacrin derivatives were synthesized and evaluated in assays for HIV replication and latency activation to establish which molecular structures must be maintained and which can tolerate changes that may be needed for better pharmacological properties. The results indicated that hydroxyl substituents at C-5 and C-20 are essential, while derivatives modified at 3-OH with aromatic esters retain anti-HIV replication and latent activation activities. The half-lives of the potent GM derivatives are over 20 h, which implies that they are stable in the plasm even though they contain ester linkages. The established structure activity relationship should be useful in the development of gnidimacrin or structurally related compounds as clinical trial candidates.

Three-, four-, and five-component host/guest crystals with multi-color and white light emission is described. Our strategy is based on the confinement of aromatic donor guests in supramolecular acceptor hosts. The supramolecular acceptor hosts (NDI-TPFB) were composed of N,N’-dipyrid-3-yl-1,4,5,8- naphthalenediimide (NDI) with two tris(pentafluorophenyl) borane (TPFB) linked by boron-nitrogen dative bonds as Lewis acid-base pairs, which spontaneously formed upon mixing the components. In the first part, a set of three-component crystals with 14 different aromatic guests were characterized to elucidate the structure-property relationships. In the latter part, a series of 17 types of four-and fivecomponent crystals were formed with the use of binary or ternary guest inclusion systems, and their structural and photophysical properties were investigated. Among them, 14 types of crystals were formed effectively without destroying the crystal structure, as determined by X-ray diffraction and fluorescence microscopy. Notably, flexible color tuning, including white light emission, was realized by tuning the guest ratio and the combinations. Various intermolecular interactions such as C-H center dot center dot center dot F interactions, pi-pi stacking, charge-transfer interactions, and inclusion phenomena were important for forming the crystals. This approach that yields a rational solution of multicomponent crystals could be potentially useful for obtaining novel photofunctional solid-state systems.

An iron (II) tetramethylated tetrapyridinoporphyrazine (FeII(2,3-tmtppa)) grafted onto sulfonated SBA-15 (SBA15@n-Pr-SO3@Fetmtppa)) has been synthesized. The catalyst was characterized using X-ray diffraction, nitrogen adsorption?desorption measurements, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, transmission electron microscopy, thermogravimetric analysis and scanning electron microscopy. XRD, XPS and ICP analyses confirmed the incorporation of iron (II) ion onto the surface of functionalized support. TGA and BET diagrams, FESEM and TEM images showed the grafting of (FeII(2,3-tmtppa)) on SBA-15 with mesoporous structure and ordered channels after immobilization process. This catalyst is able to perform C-H oxidation reaction of alkanes including toluene and ethylbenzene derivatives in the presence of molecular oxygen under the mild reaction in water. The hot filtration and reusability tests of the catalyst were studied which showed that the C-H activation reaction proceeds via heterogeneous pathway and catalyst is recoverable for at least 5 times.

Cellulose benzoate propionates (CBzP) were prepared via a staged addition of Pr2O followed by Bz(2)O to cellulose dissolved in tributylmethylammonium dimethyl phosphate (TBMADMP) providing regioselectively substituted cellulose esters in which the benzoate was preferentially installed at C2 and C3.
