We herein report the conventional and microscale parallel synthesis of selective inhibitors of human blood coagulation factor XIIa and thrombin exhibiting a 1,2,4-triazol-5-amine scaffold. Structural variations of this scaffold allowed identifying derivative 21i, a potent 29 nM inhibitor of FXIIa, with improved selectivity over other tested serine proteases and also finding compound 21m with 27 nM inhibitory activity toward thrombin. For the first time, acylated 1,2,4-triazol-5-amines were proved to have anticoagulant properties and the ability to affect thrombin- and cancer-cell-induced platelet aggregation. Performed mass spectrometric analysis and molecular modeling allowed us to discover previously unknown interactions between the synthesized inhibitors and the active site of FXIIa, which uncovered the mechanistic details of FXIIa inhibition. Synthesized compounds represent a promising starting point for the development of novel antithrombotic drugs or chemical tools for studying the role of FXIIa and thrombin in physiological and pathological processes.

The hypoxia-inducible factor-la (HIF-1 alpha) pathway has been implicated in tumor angiogenesis, growth, and metastasis. Therefore, the inhibition of this pathway is an important therapeutic target for the treatment of various types of cancers. Here, we designed and synthesized 31 ursolic acid (UA) derivatives containing a tetrazole moiety and evaluated them for their potential anti-tumor activities as HIF-la transcriptional inhibitors. Of these, compound 14d (IC50 0.8 +/- 0.2 mu M) displayed the most potent activity and compounds 14a (IC50 4.7 +/- 0.2 mu M) exhibited the most promising biological profile. Analysis of the structure-activity relationships of these compounds with HIF-1 alpha suggested that the presence of a tetrazole group located at C-28 of the UA derivatives was critical for their inhibitory activities.

Cross-coupling reactions have developed into powerful approaches for carbon-carbon bond formation. In this work, a Ni-catalyzed migratory Suzuki-Miyaura cross-coupling featuring high benzylic or allylic selectivity has been developed. With this method, unactivated alkyl electrophiles and aryl or vinyl boronic acids can be efficiently transferred to diarylalkane or allylbenzene derivatives under mild conditions. Importantly, unactivated alkyl chlorides can also be successfully used as the coupling partners. To demonstrate the applicability of this method, we showcase that this strategy can serve as a platform for the synthesis of terminal, partially deuterium-labeled molecules from readily accessible starting materials. Experimental studies suggest that migratory cross-coupling products are generated from Ni(0/II) catalytic cycle. Theoretical calculations indicate that the chain-walking occurs at a neutral nickel complex rather than a cationic one. In addition, the original-site cross-coupling products can be obtained by alternating the ligand, wherein the formation of the products has been rationalized by a radical chain process.

We successfully demonstrated that Friedel-Crafts-type allylation of phenol derivatives and allylic phosphates is catalyzed by the AgTFA/trimethylsilyl cyanide (Me3SiCN) and Pd(OAc)(2)/Me3SiCN combined systems to afford the C-allylated product in a highly regioselective manner. The corresponding silyl cyanometallates generated in situ are proposed to be the active catalytic species. Lewis acidity of the reversibly formed ion pairs is appropriately regulated for this reaction. The para-allylated anisole and phenol derivatives are selectively obtained. The para-substituted ones are converted to the ortho-allylated products. The reactivity of the catalytic systems is strongly dependent on the electronic nature of both electrophile and nucleophile. Substitution of an aromatic ring on the allylic phosphate is essential for the reaction. Thus, the competitive reaction of a 1 : 1 mixture of cinnamyl and simple allyl phosphates affords only the cinnamyl-substituted product.

The new pyridyl-substituted 3H-naphtho[2,1-b]pyrans display good photochromic properties with reversible generation of photomerocyanines, which exhibit mainly orange/red hues. The absorption of electromagnetic waves by materials is usually realized by multiple mechanisms, such as interface polarization, defect polarization, multiple reflections and scattering, conductive loss and magnetic loss. Therefore, attributing microwave absorption (MA) to specific mechanisms without separating them through effective means is not rigorous. Herein, three different structures: ZnO-C core-shell nanowires, ZnO-C wire-in-tubes and carbon nanotubes, were prepared through the molecular layer deposition of polyimide on ZnO nanowires and subsequent calcination at specific temperatures. Results show that the hollow structure is beneficial to MA performance. The reflection loss (RL) values of wire-in-tube and nanotube structures are much higher than the RL values of the core-shell structure. The RL peak of nanotube structure can reach -64.42 dB. Then again, multicomponent modulation is conducive to impedance matching resulting in board effective absorption bandwidth (EAB). The EAB of the wire-in-tube structure with two components is wider than that of the nanotube structure with one component, reaching as wide as 5.76 GHz. The results substantiate the effect of hollow structure and composition on the MA performance, providing a strong basis for the design and optimization of MA materials. (C) 2020 Elsevier Ltd.
