The development of a novel chemoselective functionalization can diversify the strategy for synthesizing the target molecules. The perfect chemoselectivity between aromatic and aliphatic aldehydes is difficult to achieve by the previous methods. The aromatic aldehyde-selective nucleophilic addition in the presence of aliphatic aldehydes was newly accomplished. Namely, the aromatic aldehyde-selective nucleophilic addition using arenes and allyl silanes proceeded in the presence of trialkylsilyl triflate and 2,2′-bipyridyl, while the aliphatic aldehydes completely remained unchanged. The reactive pyridinium-type salt intermediate derived from an aromatic aldehyde chemoselectively underwent the nucleophilic substitution. Moreover, the aromatic acetals as the protected aldehydes could be directly transformed into similar pyridinium salt intermediates, which reacted with various nucleophiles coexisting with the aliphatic aldehydes.

The title compound, N-(2-hydroxy-1,1-dimethylethyl)-3-methylbenzamide was synthesized by reacting 3-methylbenzoyl chloride or 3-methylbenzoic acid with 2-amino-2-methyl-1-propanol. In the present report, the synthesized target compound was fully characterized by various spectroscopic methods (H-1 NMR, C-13 NMR, IR, GC-MS), its composition confirmed by elemental analysis, and its structure determined and confirmed by X-ray analysis. The importance of this compound lies in its possession of an N,O-bidentate directing group. Such a structural motif is potentially suitable for metal-catalyzed C-H bond functionalization reactions.

A novel series of pyrazolo[1,5-a]pyrimidines were synthesized and proved by their spectral and elemental analysis, some elected of the newly synthesized compounds were examined for their cytotoxic activity employing MTT assay on two cancer cell lines (Breast and Hela cancers). Compounds 5, 7e and 7i showed the higher cytotoxicity against two cancer cell lines with (IC50 = 13.91 +/- 1.4 and 22.37 +/- 1.8 mu M/L), (IC50 = 6.56 +/- 0.5 and 8.72 +/- 0.9 mu M/L) and (IC50 = 4.17 +/- 0.2 and 5.57 +/- 0.4 mu M/L) for two cancer cell lines breast and hela respectively, using doxorubicin as a reference drug. The most potent cytotoxic active compounds 5, 7e and 7i presented inhibitory activity against KDM (histone lysine demethylases) with IC50 = 4.05, 1.91 and 2.31 mu M, respectively. The most potent KDM inhibitor 7e (IC50 = 1.91 mu M) showed to cause cell cycle arrest at G2/M phase by 4 folds than control and induce total apoptotic effect by 10 folds more than control. In silico studies performed on the more potent cytotoxic active compounds 5, 7e and 7i included lipinisk’s rule of five. Moreover, molecular docking study was utilized to explore the binding mode of the most active compounds to the target enzyme (PDB-ID: 5IVE). Also, some bioinformatics studies were carried out for compounds 7e and 7i using Swiss ADME (Swiss Institute of bioinformatics 2018).

Leishmaniasis being one of the six major tropical diseases that affects nearly 0.7-1.3 million people annually, has so far limited and high toxic therapeutic options. Herein, we report the synthesis, in silico, and in vitro evaluations of novel coumarin-incorporated isatin hydrazones (Spf-1 – Spf-10) as highly potent and safe antileishmanial agents. Molecular docking was initially carried out to decipher the binding confirmation of lead molecules towards the active cavity of the target protein (Leishmanolysin gp63) of Leishmania tropica. Among all the docked compounds, only Spf-6, Spf-8, and Spf-10 showed high binding affinities due to a pattern of strong conventional hydrogen bonds and hydrophobic pi-interactions. The molecular dynamics simulations showed the stable pattern of such bonding and structure-based confirmation with a time scale of 50 ns towards the top compound (Spf-10) and protein. These analyses affirmed the high stability of the system. Three out of ten compounds evaluated for their antileishmanial activity against Leishmania tropica promastigotes and amastigotes were found to be active at micromolar concentrations (IC50 range 0.1-4.13 mu mol/L), and most importantly, they were also found to be highly biocompatible when screened for their toxicity in human erythrocytes.

This work reports the synthesis of a novel quinolin-4(3H)-one based Schiff base ligand 3-[(E)-(2,5-dimethox-yphenyl)methylidene]amino-2-methylquinazolin-4(3H)-one (DMPAQ) and its coordination complexes of the type [M(DMPAQ)(phen)]X (1a-1c), where M = Cu(II), Zn(II) and Cd(II) ions, respectively, phen = 1,10-phe-nanthroline. All the synthesized compounds were characterized using UV-Visible, elemental analysis, FT-IR, H-1 NMR, Mass spectroscopy and TGA techniques. The triclinic structure of the DMPAQ is determined by employing single crystal X-ray crystallographic analysis. The characterization results suggested that the ligand, DMPAQ is bidentate and coordinate to the metal center through the lactum oxygen and the azomethine nitrogen. The synthesized DMPAQ ligand and complexes (1a-1c) were screened for their in vitro anticancer activity against the human breast adenocarcinoma cell line, MCF-7. The complexes 1a and 1b displayed significant anticancer ac-tivity against MCF-7 cells even at lower GI50 value (GI(50) = 0.016 mu M) than the standard drug doxorubicin (GI(50) = 0.018 mu M). Further, we have performed computational DFT studies on the chemical reactivity of the ligand and the three complexes by means of Conceptual Density Functional Theory (CDFT) through the Koopmans in DFT (KID) approximation to support the experimentally obtained results.

We report the on-demand generation of hexane-soluble (2-ethylhexyl)sodium (1) from 3-(chloromethyl)heptane (2) using a sodium-packed-bed reactor under continuous flow conditions. Thus, the resulting solution of 1 is free of elemental sodium and therefore suited for a range of synthetic applications. This new procedure avoids the storage of an alkylsodium and limits the handling of metallic sodium to a minimum. (2-Ethylhexyl)sodium (1) proved to be a very useful reagent and undergoes in-line Br/Na-exchanges as well as directed sodiations. The resulting arylsodium intermediates are subsequently trapped in batch with various electrophiles such as ketones, aldehydes, Weinreb-amides, imines, allyl bromides, disulfides and alkyl iodides. A reaction scale-up of the Br/Na-exchange using an in-line electrophile quench was also reported.

Here we present a comprehensive study on the [3+2]-cycloaddition of thiocarbonyl ylides with a wide variety of alkenes and alkynes. The obtained dihydro- and tetrahydrothiophene products serve as exceptionally versatile intermediates providing access to thiophenes, dienes, dendralenes, and vic-quarternary carbon centers. The use of high-pressure conditions enables thermally unstable, sterically encumbered or moderately reactive substrates to undergo the cycloaddition under mild conditions, thereby increasing the yield by up to 58%. A series of biologically relevant 1,4-benzoxazepine and 1,4-benzodiazepine derivatives were synthesized via a one-pot three-component reaction of 2-aminophenol or ortho-phenylenediamine, dimedone, and aromatic aldehydes catalyzed by UiO-66 metal-organic framework (MOF). This methodology provides excellent yields, simple procedure, short reaction time, low-cost catalyst, and mild reaction conditions. The UiO-66 MOF, which was characterized by different spectroscopic techniques such as XRD, FT-IR, and TEM, could be recovered and reused up to eight times with no significant loss of catalytic performance.
