Solvent-free, atom-efficient, and mechanochemically activated reactions have emerged as synthetic strategy for sustainable chemistry. Herein we report a new mechanochemical approach for the amide coupling of carboxylic acids and amines, mediated by combination of (1-cyano-2-ethoxy-2-oxoethylidenaminooxy)-dimethylaminomorpholinocarbenium hexafluorophosphate (COMU) or N,N,N’,N’-tetramethylchloroformamidinium hexa-fluorophosphate (TCFH) and K2HPO4. The method delivers a range of amides in high yields (70-96%) and fast reaction rates. The reaction protocol is mild, keeps stereochemical integrity of the adjacent to carbonyl stereocenters, and streamlines isolation procedure for solid amide products. Minimal waste is generated due to the absence of bulk solvent. We show that K2HPO4 plays a dual role, acting as a base and a precursor of reactive acyl phosphate species. Amide bonds from hindered carboxylic acids and low-nucleophilic amines can be assembled within 90 minutes by using TCFH in combination with K2HPO4 or N-methylimidazole. The developed mechanochemical liquid-assisted amidation protocols were successfully applied to the challenging couplings of all six carboxylate functions of biotin[6]uril macrocycle with phenylalanine methyl ester, resulting in 80% yield of highly pure hexa-amide-biotin[6]uril. In addition, fast and high-yielding synthesis of peptides and versatile amide compounds can be performed in a safe and environmentally benign manner, as verified by green metrics.

Several pyrrolo[2,3-b]pyridine-based B-RAF inhibitors are well known and some of them are currently FDA approved as anticancer agents. Based on the structure of these FDA approved B-V600E-RAF inhibitors, two series of pyrrolo[2,3-b]pyridine scaffold were designed and synthesized in attempt to develop new potent B-V600E-RAF inhibitors. The 38 synthesized compounds were biologically evaluated for their B-V600E-RAF inhibitory effect at single dose (10 mu M). Compounds with high percent inhibition were tested to determine their IC50 over (V600E)BRAF. Compounds 34e and 35 showed the highest inhibitory effect with IC50 values of 0.085 mu M and 0.080 mu M, respectively. Headed for excessive biological evaluation, the synthesized derivatives were tested over sixty diverse human cancer cell lines. Only compound 35 emerged as a potent cytotoxic agent against different panel of human cancer cell lines.

Thirty-four seco-3’R,4’R-disubstituted-2′,2′-dimethyldihydropyrano[2,3-f]chromone (seco-DSP) derivatives were designed, synthesized and evaluated for chemo-reversal activity when combined with paclitaxel or vincristine in P-gp overexpressing A2780/T and KB-VIN drug-resistant cancer cell lines. Most of the compounds displayed moderate to significant MDR reversal activities. Compound 7e showed the most potent chemo-sensitization activity with more than 1471 reversal ratio at a concentration of 10 mu M, which was higher than verapamil (VRP) (212-fold). Unexpectedly the newly synthesized compounds did not show chemosensitization activities in a non-P-gp overexpressing cisplatin resistant human ovarian cancer cell line (A2780/CDDP), implying that the MDR reversal effects might be associated with P-gp overexpression. Moreover, the compounds did not exhibit significant anti-proliferative activities against non-tumorigenic cell lines (HUVEC, HOSEC and T29) compared to VRP at the tested concentration and might be safer than VRP. In preliminary pharmacological mechanism studies, the compounds increased accumulation of DOX and promoted P-gp ATPase activity in A2780/T cell lines. Western blot analysis indicated they did not affect the expression level of P-gp in the tested MDR cell lines. Thus, further studies on these seco-DSP derivatives are merited with the goal of developing a desirable chemosensitizer drug candidate. (C) 2020 Elsevier Masson SAS. All rights reserved.

Etoxazole is a newly registered and widely used acaricide. However, its metabolites were not fully understood and might exhibit similar or even higher toxicity than parent compound. Therefore, in this study, the metabolites of etoxazole in citrus, soil and earthworms were firstly identified by an ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS). Four potential metabolites in citrus, 11 in soil, and 8 in earthworms were determined. These metabolites were then further structural elucidated based on the fragment pathways, and accurate mass measurement. The distributions of etoxazole and its main metabolites (M1, M2, M3, M4 and M5) which were identified as the dehydrogenation, hydrolysis, oxidation products of etoxazole (M0) were also monitored in citrus, soil and earthworms at different exposure periods. The 45 days exposure experiment showed that M0 gradually decreased in citrus and soil samples by 80% and 28% of the initial amounts, respectively. In earthworm samples, M0 accumulated in the bodies of the worms during 24 days exposure and then decreased with time. The dissipation rate of etoxazole were citrus > earthworms > soil. Concentrations of M1 and M3 in soil were found continuously increased with time during the experimental period. Moreover, the persistence of M1 in earthworm samples was also observed. Great attention should be paid to these two compounds due to their potential risks to both environmental and human health. (C) 2019 Published by Elsevier Ltd.

The reaction of zirconium salts with meso-tetra(4-carboxyphenyl)porphyrin (TCPP) in the presence of different modulators results in the formation of a diverse set of metal-organic frameworks (MOFs), each displaying distinct crystalline topologies. However, the synthesis of phase-pure crystalline frameworks remains challenging due to the concurrent formation of different polymorphs. The acidity and concentration of the modulator greatly influence the outcome of the MOF synthesis. By systematically varying these two parameters, selective framework formation can be achieved. In the present study, we aimed to elucidate the effect of modulator on the synthesis of zirconium-based TCPP MOFs. With the help of powder X-ray diffraction and scanning electron microscopy, modulator candidates and the optimal synthetic conditions yielding phase-pure PCN-222, PCN-223, and MOF-525 were identified. H-1 nuclear magnetic resonance analysis, thermogravimetric analysis, and N-2 gas sorption measurements were performed on select MOFs to gain insight into the relationship between their defectivity and modulator properties.

This article presents a detailed comprehensive investigation of the ortho fluoro- and chloro- substituted benzoic acids both, as isolated molecules and in the crystalline phase. Quantum chemical calculations performed within the density functional theory (DFT) formalism are used to investigate the potential energy landscapes of the molecules, taking into special consideration the effects of the interactions between the carboxylic group and the ortho halogen substituents, as well as the nature of these later on the structure and properties of the investigated systems. The structures of the relevant conformers of the molecules are discussed in comparative terms, and used to rationalize experimental data obtained for the compounds in the gas phase and isolated in low-temperature inert matrices. The UV-induced photofragmentation reactions of two of the compounds isolated in cryogenic inert matrices were studied as illustrative cases. The structures of the crystals reported previously in the literature are revisited and discussed also in a comparative basis. Particular emphasis is given to the analysis of the intermolecular interactions in the different crystals, using Hirshfeld surface analysis, the CE-B3LYP energy decomposition model and the HOMA index, and to their correlation with thermodynamic data.

In subsurface bioremediation, electron donor addition promotes microbial Fe(lII)-oxide mineral reduction that could change soil pore structure, release colloids, and alter soil surface properties. These processes in turn may impact bioremediation rates and the ultimate fate of contaminants. Columns packed with water-stable, Fe-oxide-rich soil aggregates were infused with acetate-containing artificial groundwater and operated for 20 d or 60 d inside an anoxic chamber. Soluble Fe(II) and soil colloids were detected in the effluent within one week after initiation of the acetate addition, demonstrating Fe(III)-bioreduction and colloid formation. Diffusible Br-, less diffusible 2,6-difluorobenzoate (DFBA), and non-diffusible silica-shelled silver nanoparticles (SSSNP) were used as tracers in transport experiments before and after the bioreduction. The transport of Br- was not influenced by the bioreduction. DFBA showed earlier breakthrough and less tailing after the bioreduction, suggesting alterations in flow paths and soil surface chemistry during the 20-d bioreduction treatment. Similarly, the bioreduction increased the transport of SSSNP very significantly, with mass recovery increasing from 1.7% to 25.1%. Unexpectedly, the SSSNP was completely retained in the columns when the acetate injection was extended from 20 to 60 d, while the mass recovery of DFBA decreased from 89.1% to 84.1% and Br showed no change. The large change in the transport of SSSNP was attributed to soil aggregate breakdown and colloid release (causing mechanical straining of SSSNP) and the exposure of iron oxide surfaces previously unavailable within aggregate interiors (facilitating attachment of SSSNP).
