A simple and efficient one-pot method has been developed for the synthesis of benzochromenes (4a-k) using V-CaHAp as a heterogeneous catalyst by the condensation of aldehydes, beta-naphthol and malononitrile in ethanol as solvent at room temperature for 20 min. The reaction, with this catalyst was carried out under mild reaction conditions with very good to excellent yields (89-98%). The material can be recycled very easily and reused for at least 6 runs without substantial loss in activity, which makes this methodology environmentally benign. We achieved a feasible and cost-effective synthesis by using non-toxic materials and minimal catalyst which is easy to handle. (C) 2015 The Authors. According to the Food and Agriculture Organization (FAO), Iran is the largest pistachio (Pistacia vera) producer in the world. Nevertheless, pistachio by-products (PBs) contain a high level of phenolic and cellulose compounds, it can be used as a substrate for the preparation of solid acid catalysts. In this work, the novel pistachio hull-OxTiCl4 (-) (x) catalyst was synthesized via preparing pistachio hull, product of Kerman, as a support followed by treatment with titanium tetrachloride (TiCl4) and identified by Fourier transform infrared spectroscopy, field emission scanning electron microscopy (FE-SEM), X-ray diffraction spectroscopy (EDX), thermogravimetric analysis (TG), Brunauer-Emmett-Teller (BET) and X-ray powder diffraction (XRD). The size of the nano-pistachio hull-OxTiCl4 (-) (x) nanocatalyst was shown by a scanning electron microscope below 100 nm. The catalytic activity of the solid acid catalyst has been successfully examined in a one-pot, three-component condensation reaction of aromatic aldehydes, dimedone and kojic acid in refluxing ethanol to furnish dihydropyrano[3,2-b]chromendione derivatives. Consequently, pyran annulated heterocycles were obtained. The proposed approach has some advantages as excellent yields, mild reaction conditions, short reaction times, use of agricultural waste and eco-friendly.

The current study is concerned with the identification of lead molecules based on the bis-coumarin scaffold having selective urease inhibitory and antiglycation activities. For that purpose, bis-coumarins (1-44) were synthesized and structurally characterized by different spectroscopic techniques. Eight derivatives 4, 8-10, 14, 17, 34, and 40 demonstrated urease inhibition in the range of IC50=4.4 +/- 0.21-115.6 +/- 2.13 mu M, as compared to standard thiourea (IC50=21.3 +/- 1.3 mu M). Especially, compound 17 (IC50=4.4 +/- 0.21 mu M) was found to be five-fold more potent than the standard. Kinetic studies were also performed on compound 17 in order to identify the mechanism of inhibition. Kinetic studies revealed that compound 17 is a competitive inhibitor. Antiglycation activity was evaluated using glycation of bovine serum albumin by methylglyoxal in vitro. Compounds 2, 11-13, 16, 17, 19-22, 35, 37, and 42 showed good to moderate antiglycation activities with IC50 values of 333.63-919.72 mu M, as compared to the standard rutin (IC50=294.46 +/- 1.5 mu M). Results of both assays showed that the compounds with urease inhibitory activity did not show any antiglycation potential, and vice versa. Only compound 17 showed dual inhibition potential. All compounds were also evaluated for cytotoxicity. Compounds 17, 19, and 37 showed a weak toxicity towards 3 T3 mouse fibroblast cell line. All other compounds were found to be non-cytotoxic. Urease inhibition is an approach to treat infections caused by ureolytic bacteria whereas inhibition of glycation of proteins is a strategy to avoid late diabetic complications. Therefore, these compounds may serve as leads for further research.

A new series of quinazolinone derivatives bearing pyridine, pyrimidine, pyrazole, or pyran moieties were synthesized for the purpose of anticancer cell line evaluation. Synthesis of these derivatives was achieved by the reaction of the ketone 2 with the appropriate aldehydes in the presence of either ethylcyanoacetate or malononitrile and ammonium acetate in one-pot reaction. Chalcones 6 reacted also with hydrazine hydrate to give the corresponding pyrazolines 7 and reacted with urea or thiourea to give the 2-oxopyrimidines or the 2-thioxopyrimidines 8, respectively. Evaluation of some representative examples of the newly synthesized compounds against cancer cell lines showed promising activity as anticancer agents.

In order to explore and develop new anticancer agents, three series of 2-phenylbenzimidazoles, 15-46, were condensed under simple and mild conditions using sodium metabisulfite as an oxidation agent and another series, 47-55, were obtained via a reduction reaction using sodium borohydride. All the compounds synthesized were evaluated for their in vitro anticancer activities against three human cancer cell lines. The novel compound 38 was found to be the most potent multi cancer inhibitor against A549, MDA-MB-231, and PC3 cell lines (IC50 values 4.47, 4.68 and 5.50 mu g mL(-1), respectively). In addition, compound 40 exhibited the best IC50 value of 3.55 mu g mL(-1) against the MDA-MB-231 cell line. The results demonstrated that introducing a new substituent to compounds 37-55 could improve their antiproliferative activities.

Herein, a novel ON-OFF colorimetric and fluorometric chemosensor; 1N-allyl-2-(2, 5-dimethoxyphenyl)-4, 5-diphenyl-1H-imidazole (ADPPI), was constructed for sequential determination of Cu2+ and S2- ions in aqueous media. The interaction between chemosensor ADPPI and different metal cations was investigated using UV-VIS and fluorimetric spectroscopy. ADPPI showed a favorable and good interaction with Cu2+ ions producing blue colored solution peaked at 610 nm with blue fluorescence at lambda(em) = 447 nm. The produced complex between Cu2+ ions and ADPPI can be used as a cascade probe for detection of S2- ions. The detection limits (LODs) were 1.01 nM and 1.25 mu M for Cu2+ and S2- ions, respectively (the lowest between the family of colorimetric and fluorometric chemosensors). To further increase the applicability of the proposed method, Cu2+ and S2- ions concentrations were measured in environmental water samples. (c) 2019 Elsevier B.V. All rights reserved.

An efficient and environmentally sustainable method for the synthesis of imidazo[1,2-a]pyridine derivatives by domino A(3)-coupling reaction catalyzed by Cu(II)-ascorbate was developed in aqueous micellar media in the presence of sodium dodecyl sulfate (SDS). The catalyst, a dynamic combination of Cu(II)/Cu(I), was generated in situ in the reaction mixture by mixing CuSO4 with sodium ascorbate and aided a facile 5-exo-dig cycloisomerization of alkynes with the condensation products of 2-aminopyridines and aldehydes to afford a variety of imidazo[1,2-a]pyridines in good overall yields. A simple experimental setup, water as the green medium, and inexpensive catalyst and auxiliary are some of the merits of this protocol.

An expedient and eco-friendly synthesis of 1-aryl/heteroaryl-[1,2,4]-triazolo[4,3-a]quinoxalin-4(5H)-ones (4) has been accomplished via iodobenzene diacetate mediated oxidative intramolecular cyclization of 3-(2-(aryl/heteroarylidene)hydrazinyl)-quinoxalin-2(1H)-ones (3). Ten synthesized compounds 3 and 4 (10-40 mu g) on irradiation with UV light at lambda(max) 312 nm could lead to cleavage of supercoiled pMaxGFP DNA (Form I) into the relaxed DNA (Form II) without any additive. Further, DNA cleaving ability of triazoles was quantitatively evaluated and was found to be dependent on its structure, concentration, and strictly on photoirradiation time. Mechanistic investigations using several additives as potential inhibitors/activator revealed that the DNA photocleavage reaction involves Type-I pathway leading to formation of superoxide anion radicals (O-2(-center dot)) as the major reactive oxygen species responsible for photocleavage process.

Herein, we present the first report on TiO2 NPs based hybrid nano-reactor designed and synthesized via N-(2-amino ethyl)-3-amino propyl trimethoxy silane (AAPTMS) functionalization of graphene oxide (GO). The assynthesized nano-reactor was characterized by means of several analytical-cum-spectroscopic techniques that support the successful attempt for the synthesis of TiO2@AAPTMS-rGO. Further, the present catalyst was evaluated for its catalytic performance towards an expedient access to biologically relevant synthesis of 2,4,5-triaryl imidazoles. The functional sites of TiO2@AAPTMS-rGO direct the catalysis from the reactant to product with high atom economy, shorter reaction time and excellent yields. Moreover, the present catalyst TiO2@AAPTMS-rGO shows promising reusability for several catalytic cycles. It also provides a robust approach for 2,4,5-triaryl imidazoles with improved performance and without significant loss in catalytic activity. Further, this catalyst can be upgraded for other organic transformations leading to numerous pharmaceuticals and environment benefits.
