The deoxyfluorination of alcohols is a fundamentally important approach to access alkyl fluorides, and thus the development of shelf-stable, easy-to-handle, fluorine-economical, and highly selective deoxyfluorination reagents is highly desired. This work describes the development of a crystalline compound, N-tosyl-4-chlorobenzenesulfonimidoyl fluoride (SulfoxFluor), as a novel deoxyfluorination reagent that possesses all of the aforementioned merits, which is rare in the arena of deoxyfluorination. Endowed by the multi-dimensional modulating ability of the sulfonimidoyl group, SulfoxFluor is superior to 2-pyridinesulfonyl fluoride (PyFluor) in fluorination rate, and is also superior to perfluorobutanesulfonyl fluoride (PBSF) in fluorine-economy. Its reaction with alcohols not only tolerates a wide range of functionalities including the more sterically hindered alcoholic hydroxyl groups, but also exhibits high fluorination/elimination selectivity. Because SulfoxFluor can be easily prepared from inexpensive materials and can be safely handled without special techniques, it promises to serve as a practical deoxyfluorination reagent for the synthesis of various alkyl fluorides.

Acylphosphonates having the 5,5-dimethyl-1,3,2-dioxophosphinanyl skeleton are developed as efficient intermolecular radical acylation reagents, which enable the cobalt-catalyzed Markovnikov hydroacylation of unactivated alkenes at room temperature under mild conditions. We describe the preparation of two monomers that bear complementary nucleobases at the edges (guanine-2′-deoxycytidine and 2-aminoadenine-2′-deoxyuridine) and that are conveniently protected and activated for solid-phase automated DNA synthesis. Fucosylated chondroitin sulfates (FuCSs) are a unique type of polysaccharides occurring in sea cucumber that show a variety of biological activities. In particular, well-defined FuCS oligosaccharides, consisting of a trisaccharide repeating unit of beta-D-GalNAc(4,6-diS)-(1 -> 4)-[alpha-L-Fuc(2,4-diS)-(1 -> 3)]-beta-D-GlcUA, display potent anticoagulant activity via selective inhibition of the intrinsic tenase, which could be developed into anticoagulant drugs without bleeding risk. Herein, we report an effective approach to the synthesis of FuCS oligosaccharides, as demonstrated by the successful elaboration of FuCS tri-, hexa-, and nonasaccharides. The syntheses employ an orthogonally protected trisaccharide as a pivotal building block that can be readily converted into the donor and acceptor for glycosidic coupling. In addition, the internal patterns of protecting groups, involving N-trichloroacetyl for N-acetyl group, benzylidene and benzyl groups for sulfonated hydroxyl groups, and benzoyl and methyl esters for free hydroxyl and carboxylic acid, respectively, ensure stereoselective formation of the glycosidic linkages and sequential transformation into the desired FuCS oligosaccharides.

The hypoxia-inducible factor-la (HIF-1 alpha) pathway has been implicated in tumor angiogenesis, growth, and metastasis. Therefore, the inhibition of this pathway is an important therapeutic target for the treatment of various types of cancers. Here, we designed and synthesized 31 ursolic acid (UA) derivatives containing a tetrazole moiety and evaluated them for their potential anti-tumor activities as HIF-la transcriptional inhibitors. Of these, compound 14d (IC50 0.8 +/- 0.2 mu M) displayed the most potent activity and compounds 14a (IC50 4.7 +/- 0.2 mu M) exhibited the most promising biological profile. Analysis of the structure-activity relationships of these compounds with HIF-1 alpha suggested that the presence of a tetrazole group located at C-28 of the UA derivatives was critical for their inhibitory activities.

Transition metal-catalyzed cross-couplings have been widely employed in the synthesis of many important molecules in synthetic chemistry for the construction of diverse C-C bonds. Self-assembling peptides and oligonucleotides have given rise to synthetic materials with several applications in nanotechnology. Aggregation of synthetic oligosaccharides into well-defined architectures has not been reported even though natural polysaccharides, such as cellulose and chitin, are key structural components of biomaterials. Here, we report that six synthetic oligosaccharides, ranging from dimers to hexamers, self-assemble into nanostructures of varying morphologies and emit within the visible spectrum in an excitation-dependent manner. Well-defined differences in chain length, monomer modification, and aggregation methods yield glycomaterials with distinct shapes and properties. The excitation-dependent fluorescence in a broad range within the visible spectrum illustrates their potential for use in optical devices and imaging applications. We anticipate that our systematic approach of studying well-defined synthetic oligosaccharides will form the foundation of our understanding of carbohydrate interactions in nature.

A one-pot, multicomponent-type, solvent-free Pd(II)-catalyzed direct beta-C-H activation/arylation of carboxamides involving anhydrides as substrates via in situ installation of directing group (DG) is reported. Typically, the DG-assisted beta-C-H activation/arylation of carboxamides is a two-step process comprising the installation of DG and Pd(II)-catalyzed C-H arylation. We attempted a multicomponent-type reaction comprising an anhydride, a DG (e.g. 8-aminoquinoline), an aryl iodide in the presence of the Pd(II) catalyst and an appropriate additive. Different anhydrides, DGs, aryl iodides, catalysts and additives were screened to reveal the scope of this multicomponent-type C-H arylation reaction process and various beta-C-H arylated carboxamides were obtained in satisfactory to good yields.
