Fucosylated chondroitin sulfates (FuCSs) are a unique type of polysaccharides occurring in sea cucumber that show a variety of biological activities. In particular, well-defined FuCS oligosaccharides, consisting of a trisaccharide repeating unit of beta-D-GalNAc(4,6-diS)-(1 -> 4)-[alpha-L-Fuc(2,4-diS)-(1 -> 3)]-beta-D-GlcUA, display potent anticoagulant activity via selective inhibition of the intrinsic tenase, which could be developed into anticoagulant drugs without bleeding risk. Herein, we report an effective approach to the synthesis of FuCS oligosaccharides, as demonstrated by the successful elaboration of FuCS tri-, hexa-, and nonasaccharides. The syntheses employ an orthogonally protected trisaccharide as a pivotal building block that can be readily converted into the donor and acceptor for glycosidic coupling. In addition, the internal patterns of protecting groups, involving N-trichloroacetyl for N-acetyl group, benzylidene and benzyl groups for sulfonated hydroxyl groups, and benzoyl and methyl esters for free hydroxyl and carboxylic acid, respectively, ensure stereoselective formation of the glycosidic linkages and sequential transformation into the desired FuCS oligosaccharides.

An electrosynthesis method for amide bond formation has been developed in an attempt to increase the atom economy for this class of reactions. This anion pool method electrochemically generates strong nucleophiles from amine substrates. The amine nucleophiles then react with acid anhydrides to generate amides, and the by- product from this reaction undergoes further chemical transformations to generate pharmaceutically relevant benzoic esters. These one- pot reactions are operationally simple, are performed at room temperature, and avoid rare transition metals and added bases. The amide synthesis is amenable to primary and secondary amines and a variety of anhydrides with yields up to 90% obtained. Atom economy and process mass index ( PMI) values calculated for this procedure indicate that this process can be considered greener compared to traditional amide synthesis routes used by industry. Furthermore, this electrochemical approach showed unique selectivity when substrates that contained two inequivalent amine moieties were examined.

This study aimed to elucidate the intrinsic mechanisms of PS activation by carbon nanotubes (CNTs). Singlet oxygen generation (O-1(2)) and direct CNTs-mediated electron transfer were hypothesized to be two major pathways of the oxidation of 2,4-dichlorophenol (2,4-DCP) by PS in the presence of both unmodified and modified CNTs. For the first time, roles of CNT active sites responsible for PS activation were determined using CNT derivatization and structural characterization. By selectively deactivating the carbonyl, hydroxyl or carboxylic groups on CNTs surface and linear sweep voltammetry (LSV) analysis, -C=0 groups were determined to be the main active sites contributing to the direct electron transfer oxidation, while singlet oxygen was generated at CNTs defects. Subsequent UV irradiation was shown to cause the recovery of surface defects with I-D/I-G of CNTs increasing by 21%. This resulted in the regeneration of the performance for the coupled system and allowed for multi-cycle activation of PS by CNTs. These results suggest that CNTs/PS system combined with regeneration based on UV irradiation can be used as an effective alternative process for continuous degradation of recalcitrant aqueous contaminants through the non-radical mechanism. We described the chemical synthesis of a sulfated trisaccharide repeating unit of fucosylated chondroitin sulfate (FCS), which has significant anticoagulant activity. Well-functionalized monosaccharides were readily prepared, and highly efficient glycosylations using a common activator (NIS/ TfOH) were also presented. The synthesized trisaccharide 4 could be used to extend oligosaccharide sequences.

Separation of bovine serum albumin (BSA) and hemoglobin (Hb), two proteins with almost identical molecular weight, has been a big challenge for more than two decades. Using traditional ultrafiltraion membranes separation of these proteins is possible only at the isoelectric point of one protein via electrostatic repulsion between the membrane and the other protein. Here we introduce an integral asymmetric isoporous membrane from polystyrene-block-poly(2-hydroxyethyl meth-acrylate-ran-2-(succinyloxy)ethyl methacrylate) (PS-b-P(HEMA-r-SEMA)) having random-OH and-COOH groups along the pore walls prepared by a method which combines solvent induced self-assembly of the block copolymer and nonsolvent induced phase separation (SNIPS). The membrane consists of soft isoporous channels due to swelling of the P(HEMA-r-SEMA) blocks in a hydrated state. The effective pore size of the membrane in a hydrated state is significantly lower compared to that in a dry state. These soft channels allow the permeation of BSA while retaining Hb at constant pH 7.4 where both proteins are negatively charged. In comparison to 22 commercial and in-house prepared membranes the PS-b-P(HEMA-r-SEMA) membrane presents unprecedented ideal selectivity of BSA over Hb (psi(BSA/Hb) = 16). Unlike the conventional technique in this case the electroneutrality of one protein is not mandatory which will provide significantly easier handling.

Platelet thrombosis is the main pathogeny resulting in the low curability of ischemic stroke, a leading cause of mortality and disability worldwide. Metformin, a biguanide derivative that is the first-line oral medicine for type 2 diabetes, alleviates the severity of ischemic stroke in diabetic patients and suppresses platelet activation in experimental animal model. However, the clinical implementation of commercial biguanide analogs for stroke related to platelet thrombosis remains challenging due to its weak potency, poor pharmacokinetic characteristics and possible hypoglycemia. Here, twenty-three biguanide derivatives were designed and synthesized based on the principles of bioisosteres. These derivatives were evaluated for the activity of antiplatelet thrombosis in vivo. We found that N-tri-fluoromethanesulfonyl biguanide derivative, compound b10, uniquely prevented cerebral infarction as well as neuronal function injury, and significantly decrease the mortality rate of ischemic stroke in the middle cerebral artery occlusion mice without significant side effects. We verified that b10 directly inhibited platelets thrombus formation and decreased the compactness of stroke thrombi. Particularly, b10 exhibited good potency to inhibit human platelet activation including platelet aggregation, adhesion, pseudopodia formation, integrin GPIIb/IIIa activation, CD62P expression and clot retraction. Meanwhile, the pharmacokinetics assessment showed that b10 had satisfying pharmacological characteristics including a longer duration and a higher oral absorption ratio than its parent compound. In addition, b10 remarkably ameliorated not only stroke related to platelet thrombosis but also carotid artery thrombus formation. It is concluded that the novel potent antiplatelet thrombotic agent derived from biguanide is a promising candidate for stroke treatment. (C) 2020 Elsevier Masson SAS. All rights reserved.

We synthesized N-benzoyl-protected peptide nucleic acid (PNA) monomers, which are robust under the conditions for deprotecting the 9-fluorenylmethoxycarbonyl (Fmoc) group by piperidine but are removable by aqueous ammonia and hence totally compatible with Fmoc-solid phase synthesis. This new invention expands the range of available nucleobase sequences, allowing us to use acid-sensitive PNA oligomers and purine nucleotides (both of which are difficult to use in the conventional methods) in the preparation of PNA-DNA chimeras to avoid the drawbacks of traditional PNAs.
