Because of their stable orientations and their minimal interference with native DNA interactions and folding, emissive isomorphic nucleoside analogues are versatile tools for the accurate analysis of DNA structural heterogeneity. Here, we report on a bifunctional trifluoromethylphenylpyrrolocytidine derivative ((FP)dC) that displays an unprecedented quantum yield and highly sensitive F-19 NMR signal. This is the first report of a cytosine-based dual-purpose probe for both fluorescence and F-19 NMR spectroscopic DNA analysis. (FP)dC and (FP)dC-containing DNA were synthesized and characterized; our robust dual probe was successfully used to investigate the noncanonical DNA structure, i-motifs, through changes in fluorescence intensity and F-19 chemical shift in response to i-motif formation. The utility of (FP)dC was exemplified through reversible fluorescence switching of an (FP)dC-containing i-motif oligonucleotide in the presence of Ag(i) and cysteine.

This contribution describes sulfoximine (SFI) derivatives as a novel type of acceptor for efficient thermally activated delayed fluorescence (TADF) emitters. SFI presents a unique advantage by facile introduction of versatile functionalities on its secondary amine, which allows delicate manipulation of TADF properties. In a proof of principle study, three green/yellowish green TADF emitters, namely DMAC-SFIP, DMAC-SFIC and DMAC-SFIS were developed with different electron-withdrawing N-substituents based on simple donor-acceptor (D-A) typed structure. A look into the molecular conformations indicates the existence of different types of weak intramolecular interactions to suppress nonradiative decay. More importantly, effective mixing between charge transfer (CT) and locally excited (LE) triplet states is observed for DMAC-SFIP, DMAC-SFIC due to suitable electron-deficiency in the SFI segment, resulting in acceleration of the reverse intersystem crossing (RISC) rate. Consequently, these new TADF emitters render excellent organic light-emitting diode (OLED) performances with external quantum efficiencies (EQEs) up to 20.4% and small efficiency roll-off.

The depolymerization of the biopolymer lignin can give pure aromatic monomers but selective catalytic approaches remain scarce. Here, an approach was rerouted to deliver an unusual phenolic monomer. The microporous framework copper silicates CuSH-1NaK (1) and SGU-29 (2) – prepared using hydrothermal conditions – are effective heterogeneous oxidation catalysts for producing benzaldehyde (BzAld) and benzoic acid (BzAcid) from benzyl alcohol (BzOH), under mild reaction conditions (approximately atmospheric pressure, 70 degrees C). The copper silicate heterogeneous catalysts triggered a radical reaction mechanism leading to the target products; in particular, BzAcid was formed in up to 94 % yield and 100 % selectivity from BzAld. The type of oxidant and solvent influenced the catalytic performances.

Timely myocardial reperfusion salvages ischemic myocardium from infarction, whereas reperfusion itself induces cardiomyocyte death, which is called myocardial ischemia/reperfusion (MI/R) injury. Herein, beta-carboline derivative 17c was designed and synthesized with obvious myocardial protective activity for the first time. Pretreatment of 17c effectively protected the cardiomyocyte H9c2 cells from H2O2-induced lactate dehydrogenase leakage and restored the endogenous antioxidants, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). Besides, 17c effectively protected the mitochondria through decreasing the reactive oxygen species overproduction and enhancing the mitochondrial membrane potential. As a result, 17c significantly reduced the necrosis of cardiomyocytes in H2O2-induced oxidative stress, which was more potent than polydatin. In MI/R injury rats, 17c pretreatment obviously increased the levels of SOD and GSH-Px and inhibited the apoptosis of cardiomyocytes. Through this way, the size of myocardial infarction was significantly reduced after MI/R injury in vivo, better than that of polydatin, suggesting that 17c is a promising cardioprotectant for the prevention of MI/R injury.

The extensively developed ene-type enantioselective cycloisomerization of classical 1,n-enynes provides an efficient approach to chiral cyclic 1,4-dienes. In contrast, the catalytic asymmetric heteroarenyne (heteroarene-alkyne) cycloisomerization involving the dearomative transformation of endocyclic aromatic C=C bonds remains unknown. Herein, we communicate a PdH-catalyzed enantioselective heteroarenyne cycloisomerization reaction of alkyne-tethered indole substrates (formal 1,5- and 1,6-enynes). Based on this strategy, a variety of structurally diverse chiral spiro and fused indoline derivatives bearing quaternary stereocenters and exocyclic C=C bonds are afforded in moderate to excellent yields and excellent enantioselectivities (up to 98 % ee). The classical ene-type enantioselective 1,5-enyne cycloisomerization of N-vinylpropiolamides is also developed to afford chiral 2-pyrrolones in good to excellent ee values.

Platelet thrombosis is the main pathogeny resulting in the low curability of ischemic stroke, a leading cause of mortality and disability worldwide. Metformin, a biguanide derivative that is the first-line oral medicine for type 2 diabetes, alleviates the severity of ischemic stroke in diabetic patients and suppresses platelet activation in experimental animal model. However, the clinical implementation of commercial biguanide analogs for stroke related to platelet thrombosis remains challenging due to its weak potency, poor pharmacokinetic characteristics and possible hypoglycemia. Here, twenty-three biguanide derivatives were designed and synthesized based on the principles of bioisosteres. These derivatives were evaluated for the activity of antiplatelet thrombosis in vivo. We found that N-tri-fluoromethanesulfonyl biguanide derivative, compound b10, uniquely prevented cerebral infarction as well as neuronal function injury, and significantly decrease the mortality rate of ischemic stroke in the middle cerebral artery occlusion mice without significant side effects. We verified that b10 directly inhibited platelets thrombus formation and decreased the compactness of stroke thrombi. Particularly, b10 exhibited good potency to inhibit human platelet activation including platelet aggregation, adhesion, pseudopodia formation, integrin GPIIb/IIIa activation, CD62P expression and clot retraction. Meanwhile, the pharmacokinetics assessment showed that b10 had satisfying pharmacological characteristics including a longer duration and a higher oral absorption ratio than its parent compound. In addition, b10 remarkably ameliorated not only stroke related to platelet thrombosis but also carotid artery thrombus formation.
