To improve the efficiency of the photocatalytic decarbonylation of cyclopropenones, the effects of substituents on cyclopropenone were explored. A benzothiophene-substituted aminocyclopropenone exhibited significantly improved decarbonylation efficiency to produce the corresponding ynamine, which worked as a potent dehydration condensation agent. The benzothiophene derivative was applicable to the photocatalytic reaction in the presence of potential excited-state quenchers such as oxygen and anilines. The high catalyst sensitivity would be attributed to the involvement of triplet energy transfer reaction pathway, which was not observed in the reaction with previously reported aminocyclopropenones.

A series of 1-(2-aminopropyl)-5-aryl-4-acyl-3-hydroxy-3-pyrrolin-2-ones were synthesized by three-component reactions of 4-substituted 2,4-dioxobutanoic acid methyl esters with mixtures of an aromatic aldehyde and 1,2-diaminopropane. Products of their reactions with hydrazine, p-phenetidine, and acetic anhydride were obtained. The analgesic, anti-inflammatory, and antimicrobial activities and hemostatic effect of several compounds were studied.

Separation of bovine serum albumin (BSA) and hemoglobin (Hb), two proteins with almost identical molecular weight, has been a big challenge for more than two decades. Using traditional ultrafiltraion membranes separation of these proteins is possible only at the isoelectric point of one protein via electrostatic repulsion between the membrane and the other protein. Here we introduce an integral asymmetric isoporous membrane from polystyrene-block-poly(2-hydroxyethyl meth-acrylate-ran-2-(succinyloxy)ethyl methacrylate) (PS-b-P(HEMA-r-SEMA)) having random-OH and-COOH groups along the pore walls prepared by a method which combines solvent induced self-assembly of the block copolymer and nonsolvent induced phase separation (SNIPS). The membrane consists of soft isoporous channels due to swelling of the P(HEMA-r-SEMA) blocks in a hydrated state. The effective pore size of the membrane in a hydrated state is significantly lower compared to that in a dry state. These soft channels allow the permeation of BSA while retaining Hb at constant pH 7.4 where both proteins are negatively charged. In comparison to 22 commercial and in-house prepared membranes the PS-b-P(HEMA-r-SEMA) membrane presents unprecedented ideal selectivity of BSA over Hb (psi(BSA/Hb) = 16). Unlike the conventional technique in this case the electroneutrality of one protein is not mandatory which will provide significantly easier handling.

The concept of enthalpy-entropy compensation (EEC) is one of the highly debated areas of thermodynamics. The conformational change due to restricted double-bond rotation shows a classic two-site chemical exchange phenomenon and has been extensively studied. Fifty-four analogs of N,N-diethyl-m-toluamide (DEET) as a model system were synthesized to study the thermodynamics of the partial amide bond character using nuclear magnetic resonance (NMR) spectroscopy. Line-shape analysis as a function of temperature is used to estimate the chemical exchange. Eyring analysis was then used to convert the chemical exchange rates to determine the transition state enthalpy and entropy of the molecules. The experimental design follows selective variations that perturb one aspect of the molecular system and its influence on the observed thermodynamic effect. The results of the study demonstrate that amide bond resonance in analogs of DEET follows an EEC mechanism. Simple modifications made to DEET’s structural motif alter both the enthalpy and entropy of the system and were limited overall to a temperature compensation factor, T-beta = 292.20 K, 95% CI [290.66, 293.73]. We suggest EEC as a model to describe the kinetic compensation seen in chemical exchange phenomena in analogs of DEET.

A palladium-catalysed direct alkenation of electron rich arenes in the presence of K2S2O8 with an acetic acid/1,4-dioxane solvent combination has been developed. The 1,4-dioxane co-solvent dramatically influences the rate of reaction, giving selectively disubstituted alkenes, while the addition of acetylacetone ligands was shown to increase site selectivity for the alkenation of monofunctionalized arenes. The participation of these carbonyl ligands has been confirmed by ESI-MS studies, with some key in situ intermediates in the catalytic cycle identified. Platelet thrombosis is the main pathogeny resulting in the low curability of ischemic stroke, a leading cause of mortality and disability worldwide. Metformin, a biguanide derivative that is the first-line oral medicine for type 2 diabetes, alleviates the severity of ischemic stroke in diabetic patients and suppresses platelet activation in experimental animal model. However, the clinical implementation of commercial biguanide analogs for stroke related to platelet thrombosis remains challenging due to its weak potency, poor pharmacokinetic characteristics and possible hypoglycemia. Here, twenty-three biguanide derivatives were designed and synthesized based on the principles of bioisosteres. These derivatives were evaluated for the activity of antiplatelet thrombosis in vivo. We found that N-tri-fluoromethanesulfonyl biguanide derivative, compound b10, uniquely prevented cerebral infarction as well as neuronal function injury, and significantly decrease the mortality rate of ischemic stroke in the middle cerebral artery occlusion mice without significant side effects. We verified that b10 directly inhibited platelets thrombus formation and decreased the compactness of stroke thrombi. Particularly, b10 exhibited good potency to inhibit human platelet activation including platelet aggregation, adhesion, pseudopodia formation, integrin GPIIb/IIIa activation, CD62P expression and clot retraction. Meanwhile, the pharmacokinetics assessment showed that b10 had satisfying pharmacological characteristics including a longer duration and a higher oral absorption ratio than its parent compound. In addition, b10 remarkably ameliorated not only stroke related to platelet thrombosis but also carotid artery thrombus formation.
