Separation of bovine serum albumin (BSA) and hemoglobin (Hb), two proteins with almost identical molecular weight, has been a big challenge for more than two decades. Using traditional ultrafiltraion membranes separation of these proteins is possible only at the isoelectric point of one protein via electrostatic repulsion between the membrane and the other protein. Here we introduce an integral asymmetric isoporous membrane from polystyrene-block-poly(2-hydroxyethyl meth-acrylate-ran-2-(succinyloxy)ethyl methacrylate) (PS-b-P(HEMA-r-SEMA)) having random-OH and-COOH groups along the pore walls prepared by a method which combines solvent induced self-assembly of the block copolymer and nonsolvent induced phase separation (SNIPS). The membrane consists of soft isoporous channels due to swelling of the P(HEMA-r-SEMA) blocks in a hydrated state. The effective pore size of the membrane in a hydrated state is significantly lower compared to that in a dry state. These soft channels allow the permeation of BSA while retaining Hb at constant pH 7.4 where both proteins are negatively charged. In comparison to 22 commercial and in-house prepared membranes the PS-b-P(HEMA-r-SEMA) membrane presents unprecedented ideal selectivity of BSA over Hb (psi(BSA/Hb) = 16). Unlike the conventional technique in this case the electroneutrality of one protein is not mandatory which will provide significantly easier handling.

This report details a decarboxylative cross-coupling of (hetero)aryl carboxylates with iodoarenes in the presence of a gold catalyst (>25 examples, up to 96% yield). In subsurface bioremediation, electron donor addition promotes microbial Fe(lII)-oxide mineral reduction that could change soil pore structure, release colloids, and alter soil surface properties. These processes in turn may impact bioremediation rates and the ultimate fate of contaminants. Columns packed with water-stable, Fe-oxide-rich soil aggregates were infused with acetate-containing artificial groundwater and operated for 20 d or 60 d inside an anoxic chamber. Soluble Fe(II) and soil colloids were detected in the effluent within one week after initiation of the acetate addition, demonstrating Fe(III)-bioreduction and colloid formation. Diffusible Br-, less diffusible 2,6-difluorobenzoate (DFBA), and non-diffusible silica-shelled silver nanoparticles (SSSNP) were used as tracers in transport experiments before and after the bioreduction. The transport of Br- was not influenced by the bioreduction. DFBA showed earlier breakthrough and less tailing after the bioreduction, suggesting alterations in flow paths and soil surface chemistry during the 20-d bioreduction treatment. Similarly, the bioreduction increased the transport of SSSNP very significantly, with mass recovery increasing from 1.7% to 25.1%. Unexpectedly, the SSSNP was completely retained in the columns when the acetate injection was extended from 20 to 60 d, while the mass recovery of DFBA decreased from 89.1% to 84.1% and Br showed no change. The large change in the transport of SSSNP was attributed to soil aggregate breakdown and colloid release (causing mechanical straining of SSSNP) and the exposure of iron oxide surfaces previously unavailable within aggregate interiors (facilitating attachment of SSSNP). A highly enantioselective alpha-ketol rearrangement has been developed. In the presence of a chiral Cu-bisoxazoline complex, achiral beta-hydroxy-alpha-dicarbonyls were isomerized to chiral alpha-hydroxy-beta-dicarbonyls and their bicyclic derivatives in excellent yields and enantioselectivities. Enantioenriched 2-acyl-2-hydroxy cyclohexan-1-ones, dihydroxyhexahydrobenzofuranones, and dihydroxyhexahydro-cycloheptafuranones, with up to three stereocenters, were readily prepared from achiral starting materials in one operation. The reaction is applicable to the desymmetrization of meso substrates and kinetic resolution of racemic alcohols.

DNA-encoded chemical libraries are increasingly used in pharmaceutical research because they enable the rapid discovery of synthetic protein ligands. Here we explored whether target-class focused DNA-encoded chemical libraries can be cost-effective tools to achieve robust screening productivity for a series of proteins. The study revealed that a DNA-encoded library designed for NAD(+)-binding pockets (NADEL) effectively sampled the chemical binder space of enzymes with ADP-ribosyltransferase activity. The extracted information directed the synthesis of inhibitors for several enzymes including PARP15 and SIRT6. The high dissimilarity of NADEL screening fingerprints for different proteins translated into inhibitors that showed selectivity for their target. The discovery of patterns of enriched structures for six out of eight tested proteins is remarkable for a library of 58 302 DNA-tagged structures and illustrates the prospect of focused DNA-encoded libraries as economic alternatives to large library platforms.

Although methyl transfer reactions are important in both chemical and biological systems, there is a need for thermodynamic parameters related to methyl affinity and O-CH3 bond dissociation enthalpies (BDEs) relevant to a full understanding of the mechanisms of methyl transfer reactions. As a prelude to the construction of a database of O-CH3 BDEs, the present work examines the reliability of a series of theoretical methods for the prediction of O-CH3 BDEs using a set of 25 compounds that included both aromatic and non-aromatic molecules. The BDEs calculated by density functional theory (DFT) with traditional exchange-correlation functions exhibited much larger errors than those obtained by either the M06-2X or G4 methods. For the non-aromatic compounds, M06-2X/def2-TZVP performed slightly better than G4, but G4 was more accurate for the aromatic molecules. As a result, we recommend G4 as the preferred method for the theoretical estimation of O-CH3 bond dissociation enthalpies, although M06-2X may be a good alternative for large complex molecules when the use of G4 is impractical.

A palladium-catalysed direct alkenation of electron rich arenes in the presence of K2S2O8 with an acetic acid/1,4-dioxane solvent combination has been developed. The 1,4-dioxane co-solvent dramatically influences the rate of reaction, giving selectively disubstituted alkenes, while the addition of acetylacetone ligands was shown to increase site selectivity for the alkenation of monofunctionalized arenes.
