Platelet thrombosis is the main pathogeny resulting in the low curability of ischemic stroke, a leading cause of mortality and disability worldwide. Metformin, a biguanide derivative that is the first-line oral medicine for type 2 diabetes, alleviates the severity of ischemic stroke in diabetic patients and suppresses platelet activation in experimental animal model. However, the clinical implementation of commercial biguanide analogs for stroke related to platelet thrombosis remains challenging due to its weak potency, poor pharmacokinetic characteristics and possible hypoglycemia. Here, twenty-three biguanide derivatives were designed and synthesized based on the principles of bioisosteres. These derivatives were evaluated for the activity of antiplatelet thrombosis in vivo. We found that N-tri-fluoromethanesulfonyl biguanide derivative, compound b10, uniquely prevented cerebral infarction as well as neuronal function injury, and significantly decrease the mortality rate of ischemic stroke in the middle cerebral artery occlusion mice without significant side effects. We verified that b10 directly inhibited platelets thrombus formation and decreased the compactness of stroke thrombi. Particularly, b10 exhibited good potency to inhibit human platelet activation including platelet aggregation, adhesion, pseudopodia formation, integrin GPIIb/IIIa activation, CD62P expression and clot retraction. Meanwhile, the pharmacokinetics assessment showed that b10 had satisfying pharmacological characteristics including a longer duration and a higher oral absorption ratio than its parent compound. In addition, b10 remarkably ameliorated not only stroke related to platelet thrombosis but also carotid artery thrombus formation. It is concluded that the novel potent antiplatelet thrombotic agent derived from biguanide is a promising candidate for stroke treatment. (C) 2020 Elsevier Masson SAS. All rights reserved.

Eleven new tetrahydrobenzo[b]pyran derivatives were synthesized via a three component reaction of different aromatic aldehydes, methyl cyanoacetate and 1,3-cyclohexadione, with water as solvent under catalyst-free microwave irradiation. The structures of all the new molecules were well analysed and their structures established by using various spectral techniques (H-1 NMR, C-13 NMR, N-15 NMR and HRMS). Various advantages of reported protocol are the ease of preparation, short reaction times (10 min), aqueous solvent and excellent yields (89-98%). In this work, 1,3,5,7-tetraazaadamantan-1-ium chloride (AIL) functionalized silica-coated calcium oxide hybrid nanocatalyst (CaO@SiO2@AIL) as a novel, efficient, green and recyclable heterogeneous ionic liquid catalyst was synthesized. Catalytic activity of the CaO@SiO2@AIL hybrid nanoparticles was investigated for synthesis of the pharmaceutically valuable 2-imino-2H-chromene and dihydropyrano[c]chromene derivatives. A wide range of amines and aromatic aldehydes containing either electron-withdrawing or electron-donating substituent were examined using optimized conditions to produce the desired products. 2-Imino-2H-chromenes were synthesized under solvent-free condition, and dihydropyrano[c]chromenes were prepared in aqueous medium as green conditions within short reaction times, high yields and using easy workup procedures. Structure confirmation and surface properties of the core/shell hybrid nanoparticles were considered via Fourier transform infrared spectroscopy, X-ray powder diffraction, scanning electron microscope, energy-dispersive X-ray spectroscopy, WDS map scan, thermogravimetric and elemental analyses. This IL-supported heterogeneous nanocatalyst can be reused at least six times without considerable loss of its performance.

Totally colorless, transparent, and highly refractive poly(phenylene thioether ether)s (PPTEs) containing thianthrene-2,7-disulfanyl moiety were developed in this study. A new 4,4′-diol aromatic compound, 2,7-(4,4’-diphenol)thiothianthrene (DPTT), with a high sulfur content was designed and synthesized to develop polymers with high refractive indices and high thermal stabilities. The PPTEs were prepared by polycondensation of DPTT with organic dihalogen compounds, such as 2,6-difluorobenzonitrile (DFBN) and 4,6-dichloro-2-(methylthio) pyrimidine (DCMP). The obtained polymers showed good thermal stabilities such as relatively high glass transition temperatures (T(g)s) in the range of 153-176 degrees C and 5% weight loss temperatures (T-5%) over 344 degrees C. The cut-off wavelengths (lambda(0)) of the PPTE films are shorter than 343 nm, and they have a transmittance higher than 93% at 550 nm. The combination of the pyrimidine and high sulfur-containing thianthrene derivatives provides the PPTEs with a high refractive index of 1.7204 and a small birefringence of 0.0106 at 637 nm.

A trifluoroacetic-acid-mediated desulfurilative sulfonylthiolation of arenes using SS-morpholino dithiosulfonate is described. This system is based on selective activation of the morpholino group over the tosyl group of the doubly transformable sulfur surrogate. Mechanistic studies suggested that the reaction proceeds through electrophilic aromatic substitution followed by sulfur extrusion. The wide substrate scope of this reaction and the transformability of the resulting thiosulfonates enable expeditious access to divergent multifunctionalized sulfides.
