Highly enantioselective desymmetrisation of Fittig’s lactones with alcohols is promoted by bifunctional cinchona squaramides. The reactions were carried out with monodeuterated methanol to detect possible hidden racemisation of the stereogenic centre. Current evidence suggests that racemisation was not a relevant process for most substrates; partial erosion of enantioselectivity was only detected with ortho-substituted aryl derivates. The resultant glutaric acid derivatives possess a scaffold that is common in natural products and the compounds are also useful chiral building blocks for further synthetic endeavours.

A highly enantioselective alpha-ketol rearrangement has been developed. In the presence of a chiral Cu-bisoxazoline complex, achiral beta-hydroxy-alpha-dicarbonyls were isomerized to chiral alpha-hydroxy-beta-dicarbonyls and their bicyclic derivatives in excellent yields and enantioselectivities. Enantioenriched 2-acyl-2-hydroxy cyclohexan-1-ones, dihydroxyhexahydrobenzofuranones, and dihydroxyhexahydro-cycloheptafuranones, with up to three stereocenters, were readily prepared from achiral starting materials in one operation. The reaction is applicable to the desymmetrization of meso substrates and kinetic resolution of racemic alcohols.

Inorganic persulfate salts were identified as efficient reagents for the oxidative aromatization of 3,4-dihydroquinolin-2(1H)-ones through the activation of readily available transition metals, such as iron and copper. The feasible protocol conforming to the requirement of green chemistry was utilized in the preparation of the key intermediate (7-(4-chlorobutoxy)quinolin-2(1H)-one 2) of brexpiprazole in 80% isolated yield on a 100 g scale, and different quinolin-2(1H)-one derivatives with various functional groups were demonstrated in 52-89% yields.

Herein, we have introduced sulfonimidoyl azide as a precursor in a Cu-catalyzed Chan-Evans-Lam reaction with aryl boronic acid for the synthesis of N-aryl sulfonimidamide. The CuCl catalyzed reactions of sulfonimidoyl azides and aryl boronic acids have been proceeded in the presence of triethylamine (Et3N) as base in methanol at room temperature under oxygen atmosphere and resulted moderate to good yields of corresponding products in very short time. The synthetic utilities of N-aryl sulfonimidamides have been established by iodination reactions and Suzuki-Miyaura coupling reactions.

We report an efficient and straight forward access to nitrile substituted enol esters via ionic liquid [BMIM]OH mediated hydrocarboxylation of alkynylnitriles under mild conditions. This atom economical transition metal-free protocol gives an easy access to a variety of such enol esters with excellent regio and Z-stereoselectivity. Reusability of [BMIM]OH without losing of significant amount of yield is another noticeable feature of this article.

A novel palladium(II)/copper(II)-catalyzed sulfidation of the C-H bond in electron-rich arenes and in pentafluorobenzene with disulfides was developed. A series of new Olaparib derivatives was designed and synthesized, and their inhibitory activities against poly (ADP-ribose) polymerases-1 (PARP-1) enzyme and cancer cell line MDA-MB-436 in vitro were evaluated. The results showed that compound 5l exhibited the most potent inhibitory effects on PARP-1 enzyme (16.10 +/- 1.25 nM) and MDA-MB-436 cancer cell (11.62 +/- 2.15 mu M), which was close to that of Olaparib. As a PARP-1 inhibitor had been reported to be viable to neuroprotection, in order to search for new multitarget-directed ligands (MTDLs) for the treatment of Alzheimer’s disease (AD), the inhibitory activities of the synthesized compounds against the enzymes AChE (from electric eel) and BChE (from equine serum) were also tested. Compound 5l displayed moderate BChE inhibitory activity (9.16 +/- 0.91 mu M) which was stronger than neostigmine (12.01 +/- 0.45 mu M) and exhibited selectivity for BChE over AChE to some degree. Molecular docking studies indicated that 5l could bind simultaneously to the catalytic active of PARP-1, but it could not interact well with huBChE. For pursuit of PARP-1 and BChE dual-targeted inhibitors against AD, small and flexible non-polar groups introduced to the compound seemed to be conducive to improving its inhibitory potency on huBChE, while keeping phthalazine-1-one moiety unchanged which was mainly responsible for PARP-1 inhibitory activity. Our research gave a clue to search for new agents based on AChE and PARP-1 dual-inhibited activities to treat Alzheimer’s disease.
