Monoacylated symmetrical diamines are building blocks or intermediates of several well-established drugs. However, for the symmetrical diamines, the two amine groups located in a similar chemical environment make the selective monoacylation rather difficult and no kinetics study has ever been reported before. To comprehend this kind of reaction more deeply, a continuous flow microfluidic system was developed to evaluate the reaction kinetics of m-phenylenediamine and benzoic anhydride. At first, the reaction order of each reactant, pre-exponential factors and activation energies were determined in turn. The possible reaction mechanism was postulated from the reaction order. Then, a series of validation experiments were designed to show the accuracy and the applied range (concentration and temperature) of the kinetic model. Next, to obtain a controlled selectivity, the effects of initial concentration, reaction temperature and molar ratio on the selectivity were investigated through the model simulation. In the end, under the conditions optimized by the kinetic model, 96.9% selectivity and 92.5% yield were obtained in the microreactor.

We synthesized two kinds of monocarbamothioates TZO(R) containing a thiazolidin-2-one (TZO) skeleton and TZD(R) containing a thiazolidine-2,4-dione (TZD) skeleton. In the reaction of TZO(R) and phenoxypropyrenesulfide (PPS) using tetrabutylammonium chloride (TBAC) as a catalyst in 1-methyl-2-pyrrolidinone at 60 degrees C, anionic ring-opening polymerization of PPS proceeded to give the corresponding linear polysulfides, polyPPS, with M-n = 75 860-206 700 (M-w/M-n = 1.54-1.73) in high yield. In contrast, in the reaction of TZD(R) and PPS, the continuous insertion reaction of PPS into TZD(R), i.e., living ring-expansion polymerization, occurred to afford cyclic polysulfides TZD(R)-c-poly(PPS)(n) with M-n = 1870-33 420 (M-w/M-n = 1.07-1.26) in high yield. The ring size could be controlled by the feed ratio of TZD(R)/PPS in the range of 1/5-1/100.

Highly enantioselective desymmetrisation of Fittig’s lactones with alcohols is promoted by bifunctional cinchona squaramides. The reactions were carried out with monodeuterated methanol to detect possible hidden racemisation of the stereogenic centre. Current evidence suggests that racemisation was not a relevant process for most substrates; partial erosion of enantioselectivity was only detected with ortho-substituted aryl derivates. The resultant glutaric acid derivatives possess a scaffold that is common in natural products and the compounds are also useful chiral building blocks for further synthetic endeavours.

Fucosylated chondroitin sulfates (FuCSs) are a unique type of polysaccharides occurring in sea cucumber that show a variety of biological activities. In particular, well-defined FuCS oligosaccharides, consisting of a trisaccharide repeating unit of beta-D-GalNAc(4,6-diS)-(1 -> 4)-[alpha-L-Fuc(2,4-diS)-(1 -> 3)]-beta-D-GlcUA, display potent anticoagulant activity via selective inhibition of the intrinsic tenase, which could be developed into anticoagulant drugs without bleeding risk. Herein, we report an effective approach to the synthesis of FuCS oligosaccharides, as demonstrated by the successful elaboration of FuCS tri-, hexa-, and nonasaccharides. The syntheses employ an orthogonally protected trisaccharide as a pivotal building block that can be readily converted into the donor and acceptor for glycosidic coupling. In addition, the internal patterns of protecting groups, involving N-trichloroacetyl for N-acetyl group, benzylidene and benzyl groups for sulfonated hydroxyl groups, and benzoyl and methyl esters for free hydroxyl and carboxylic acid, respectively, ensure stereoselective formation of the glycosidic linkages and sequential transformation into the desired FuCS oligosaccharides.

Numerous electrophilic thiocyano oxyfunction-alization reactions of alkenes have been achieved using N-thiocyano-dibenzenesulfonimide, which is a new electrophilic thiocyanation reagent and could be easily prepared in two steps from dibenzenesulfonimide. BACKGROUND: Chemical pesticides are the main measures for pest control, but have caused growing resistance of pests and brought a series of environmental problems. Development of high-efficient insecticidal molecules with novel scaffolds is therefore particularly urgent. RESULTS: Based on a [5 + 1] annulation reaction with 5-amino-1H-phenylpyrazole and dialkyl bromomalonate, 27 novel five-substituted 4,5-dihydropyrazolo[1,5-a]quinazolines were designed following the intermediate derivatization method and synthesized. Bioassay results indicated that most of the test compounds displayed good insecticidal activities against Plutella xylostella, Spodoptera frugiperda, and Solenopsis invicta. In particular, the insecticidal activities of compounds 4a, 4f, and 4m against P. xylostella [median lethal concentration (LC50) values ranged from 3.87 to 5.10 mg L-1] were comparable to that of indoxacarb (LC50 = 4.82 mg L-1). In addition, compounds 4a and 9e showed similar high insecticidal activities against Spodoptera frugiperda (mortality rate = 79.63% and 72.12%) at 100 mg L-1, comparable to that of fipronil (mortality rate: 68.44%); compound 9a showed possible delayed toxicity against Solenopsis invicta (mortality rate: 95.66%) after 5 days of treatment at 1.0 mg L-1. CONCLUSION: Due to their high insecticidal activities against P. xylostella, compound 4m, 4a, and 4f could be considered as qualified candidates for novel insecticide. Several other 4,5-dihydropyrazolo[1,5-a]quinazolines with relatively high bioactivity, such as compounds 9a and 9e, are also worth further optimization as potential insecticide or anticide candidates.

Herein we describe the development of a focused series of functionalized pyridazin-3(2H)-one-based formyl peptide receptor (FPR) agonists that demonstrate high potency and biased agonism. The compounds described demonstrated biased activation of prosurvival signaling, ERK1/2 phosphorylation, through diminution of the detrimental FPR1/2-mediated intracellular calcium (Ca-i(2+)) mobilization. Compound 50 showed an EC50 of 0.083 mu M for phosphorylation of ERK1/2 and an approximate 20-fold bias away from Ca-i(2+) mobilization at the hFPR1.
