ANTIPOLIOVIRUS ACTIVITY OF ISOTHIAZOLE DERIVATIVES – MODE OF ACTION OF 5,5′-DIPHENYL-3,3′-DIISOTHIAZOLE DISULFIDE (DID)

In this report we describe the antiviral activity of 5,5′-diphenyl-3,3’diisothiazole disulfide (DID) and discuss its mode of action. DID selectively inhibits the replication of poliovirus type 1 (therapeutic index = 255) by affecting some early process of the virus growth cycle. The compound does not interfere with adsorption and internalization of virus to HEp-2 cells, nor with uncoating of the viral RNA. However, no viral RNA synthesis occurs after 2 h postinfection in the presence of 50 mu M DID. Thus, we investigated some molecular events in poliovirus replication occurring between uncoating and viral RNA synthesis. In our experimental design, we studied the activity of RNA polymerase complex isolated from HEp-2 infected cells in the presence or absence of DID. Our results showed that the RNA polymerase complex was formed in the presence of DID, On the contrary, DID markedly inhibited poliovirus RNA synthesis in a cell-free system using RNA polymerase complex isolated from infected cells. These findings indicate that DID may exert its antiviral activity by preventing viral RNA chain elongation via the inhibition of replicase activity and/or interfering with viral RNA polymerase complex.

We have presented the synthesis of novel thiazolo-and isothiazolo-apomorphines 12-17 resulting-in part-from an unexpected isomerization step occurred during the acid-catalyzed rearrangement of precursor thiazolo-morphinandienes 3-5. These 2,3-disubstituted apomorphines represent a new group of A-ring substituted aporphines. The receptor binding studies revealed that with the exception of two derivatives all the tested compounds have limited affinity for dopamine-receptor subtypes. Functional calcium assay for the most active isothiazolo-apomorphine showed higher affinities for D-1 and D-2L subtypes. The docking of these ligands has been modelled to human D-2 and D-3 receptors. NAD(P)H Quinone Oxidoreductase 1 (NQO1)-Bioactivated Pronqodine A Regulates Prostaglandin Release from Human Synovial Sarcoma Cells

Natural products have contributed to the elucidation of biological mechanisms as well as drug discovery research. Even now, the expectation for natural products is undiminished. We screened prostaglandin release inhibitors that had no effect on in vitro cyclooxygenase activity derived from natural product sources and discovered pronqodine A. Evaluation of the biological activity of pronqodine A revealed that the NAD(P)H dehydrogenase quinone 1 (NQO1) converted pronqodine A into a two-electron reductive form. The reductive form underwent autoxidation and reversed to its native form immediately with the generation of reactive oxygen species. Further investigations proved that pronqodine A inhibited cyclooxygenase enzyme activity only in the presence of NQO1. Pronqodine A acts as a potential bioreductive compound, inhibiting prostaglandin release in selectively activated NQO1-expressing cells.

Several new derivatives of 5-hydrazino-3-methyl-4-isothiazolecarboxylic ethyl esters were synthesized. Using 4-aminoacetophenone, the hydrazine group was transformed in position 5 in the hydrazone which reacted with the isocyanates, aldehydes and sugars. Thirteen newly synthesized compounds were tested for their ability to affects the immunological response in vitro in several rodent models. The immunoregulatory properties of the compounds were, differential and dose-dependent, The strongest activity was exhibited by 5-{N’-[1-4}-4-[3-(-methoxyphenyl)-ureido]-phenylethylidene]-hydrazino}-3-methyl-4-isothiazolecarboxylic acid ethyl ester (compound 3a). The compound strongly inhibited the secondary, humoral immune response to sheep erythrocytes and the proliferative response of mouse splenocytes to concanavalin A and pokeweed mitogen. The immunotropic activities of the new isothiazole derivatives and potential application of the compounds in therapy are discussed.

CONDENSATION OF 5-AMINOISOTHIAZOLES WITH N-METHYL-IMIDOYL CHLORIDE – RING-TRANSFORMATION WITH PARTICIPATION OF 10-S-3-TYPE SULFURANE

In the presence of 1,4-diazabicyclo[2.2.2]octane, 5-amino-3-methylisothiazole (4a) in acetonitrile condensed with N-methyl-p-chlorobenzimidoyl chloride (5a) to afford two kinds of compounds along with several minor products. One of the two was 1 : 1 condensed product (6a) and the other was 2 : 1 condensed product (7a). The latter compound was obtained in higher yield when an excess 4a was treated with 5a. On the other hand, in the absence of the additional base, 2 : 3 condensed compound (8a) was isolated as a major product besides 6a and 7a. 8a was obtained by treatment of 7a with 2 equiv. of imidoyl chloride in 21% yield. Reaction of 5-amino-3-phenylisothiazole (4b) with imidoyl chloride (5a,b) furnished the analogous products. On the other hand, condensation of 5-amino-3-phenylisoxazole (4c) with 5a furnished the oxygen analogue (6d) corresponding to 6b as a sole isolated product. The formation of 7 is explained by ring-transformation from isothiazole into thiadiazole via hypervalent 10-S-3 type sulfurane (B). According to a variety of investigations of heterocycles containing sulfur atom, it has been suggested that 10-S-3 type sulfurane is an important intermediate in several reactions.1,2. In a previous paper, we reported that a ring transformed thiadiazole derivative (1) instead of the expected isothiazole derivative (2) was isolated as a sole product in the reaction of 5-amino-3-methylisothiazole with arylnitrile.3a The fact was explained by invoking 10-S-3 sulfurane (A) as a intermediate. In connection with this finding, we describe in this paper results of the reaction of 5-aminoisoazole derivatives (4a-c) with imidoyl chloride (5a,b) under several conditions.

Synthesis and experimental versus theoretical research on spectroscopic and electronic properties of 3-methyl-4-nitroisothiazole

The alternative way of synthesis of valuable 5-amino-3-methylisothiazole and 3-methyl-4-nitroisothiazole has been presented and described. The stability of molecules of 3-methyl-4-nitroisothiazole arising from hyper conjugative interaction has been estimated using natural bond orbital (NBO 6.0) analysis. The combined frontier molecular orbital and NBO analysis of canonical molecular orbitals was used to predict the most probable sites of 3-methyl-4-nitroisothiazole molecule where single electron can be accepted to and detached from. Measured reduction potential of 3-methyl-4-nitroisothiazole and calculated electron affinity point out that this compound is susceptible to reduction at the similar level of 1-methyl-3-nitropyrazole and this susceptibility is much more than respective 1-methyl-4-nitropyrazole. UV-Vis spectra analysis reveals a nature of valence electron excitation and electron transition of 3-methyl-4-nitroisothiazole. Besides, unambiguous assignment of NMR signal shifts of carbon atoms of isothiazole ring of 3-methyl-4-nitroisothiazole, 5-amino-3-methylisothiazole and intermediate and side products of synthesis was conducted thanks to full detailed analysis of H-1, C-13 NMR spectra and their two dimensional (2D) variants.
