Synthesis, Bioactivity and Crystal Structure Analysis of Novel Benzo[d]isothiazol-3(2H)-ones

Two compounds, 3-oxo-N-o-tolylbenzo[d]isothiazole-2(3H)-carboxamide (1) and N-(2-methoxyphenyl)-3-oxobenzo[d]isothiazole-2(3H)-carboxamide (2), were synthesized from the initial compound benzo[d]isothiazol-3(2H)-one (BIT) and characterized by H-1 NMR, IR and elemental analysis, respectively. The single crystals of compounds 1 and 2 were obtained and determined by X-ray diffraction analysis. The preliminary results of biological activity experiment show that some of the title compounds exhibited a favorable antimicrobial activity.

In vitro and in vivo anticryptococcal activities of a new pyrazolo-isothiazole derivative

We investigated the activity of a pyrazolo-isothiazole derivative (G8) against Cryptococcus neoformans. A first screening test showed that G8 at 10 mg/L inhibited the growth of 14 of 15 clinical isolates tested. Killing experiments showed that fungicidal activity was achieved after 8 h of treatment with G8 at concentrations greater than or equal to10 mg/L. In a murine model of systemic cryptococcosis, G8 was effective at prolonging survival compared with the controls. Our data indicate that this new derivative has a potential therapeutic role in infections caused by C. neoformans.

[5-H-3]Uridine incorporation into RNA showed that the compounds tested reduced pollovirus RNA synthesis, which was completely shut off after 2 h of incubation and reduced by 50-60% after 4 h. Also, pretreatment of the cell cultures with the compounds for 24 h caused a substantial inhibition of viral replication. The data suggest that the four isothiazole derivatives may have a multi-step antiviral mode of action different from their iminodithiole precursors.

The chalcogen bond, the noncovalent, electrostatic attraction between covalently bonded atoms in group 16 and Lewis bases, is present in protein-ligand interactions based on X-ray structures deposited in the Protein Data Bank (PDB). Discovering protein-ligand chalcogen bonding in the PDB employed a strategy that focused on searching the database for protein complexes of five-membered, heterocyclic ligands containing endocyclic sulfur with endo electron-withdrawing groups (isothiazoles; thiazoles; 1,2,3-, 1,2.4-, 1,2,5-, 1,3,4-thiadiazoles) and thiophenes with exo electron-withdrawing groups, e.g., 2-chloro, 2-bromo, 2-amino, 2-alkylthio. Out of 930 ligands investigated, 33 or 3.5% have protein-ligand S-O interactions of which 31 are chalcogen bonds and two appear to be S-HO hydrogen bonds. The bond angles for some of the chalcogen bonds found in the PDB are less than 90 degrees, and an electrostatic model is proposed to explain this phenomenon.

The reactivity of the 4-amino-2,3-dihydro-1H-lambda 1(6)-isothiazole-1,1-dioxide (beta-amino-gamma-sultam) framework has not been studied sufficiently. Here we describe the chemical properties of this heterocyclic system toward electrophiles on spiranic and non-spiranic substrates. A variety of C-electrophiles (acetic anhydride, benzoyl chloride, DMFDMA, 4,4-dimethoxybutan-2-one) and heteroatom electrophiles (bromine, nitrosyl acetate) have been explored. Both the C-5 and 4-amino positions of the beta-amino-gamma-sultam system are able to undergo electrophilic reactions. Heteroatom electrophiles attack the C-5 position, whereas carbo-electrophiles affect the amino group. beta-Amino-gamma-sultams also were used as starting compounds for the synthesis of 6- or 7-substituted 1 lambda(6)-isothiazolo[4,5-b]pyridine-1,1-dioxides through condensation reaction and palladium-catalyzed oxidative coupling.
