A metal-free aromatic electrophilic substitution reaction using nitrosoarenes as the electrophile in 1,1,1,3,3,3-hexafluoroisopropanol (HFIP) is reported. HFIP activates nitrosoarene towards the electrophilic C-H amination followed by a concomitant N-O bond cleavage to deliver diarylamines without requiring any extra reagent or further treatment. The dual electrophilic nature of nitrosoarene permits a switchover of selectivity to C-H oxygenation furnishing dearomatizedp-iminoquinones following a unique mechanistic rationale of two consecutive [2,3] sigmatropic rearrangement in nitroso-chemistry.

Solvent reorganization is a major driving force of protein-ligand association, but the contribution of binding site waters to ligand affinity is poorly understood. We investigated how altered interactions with a water network can influence ligand binding to a receptor. A series of ligands of the A(2A) adenosine receptor, which either interacted with or displaced an ordered binding site water, were studied experimentally and by molecular dynamics simulations. An analog of the endogenous ligand that was unable to hydrogen bond to the ordered water lost affinity and this activity cliff was captured by molecular dynamics simulations. Two compounds designed to displace the ordered water from the binding site were then synthesized and evaluated experimentally, leading to the discovery of an A(2A) agonist with nanomolar activity. Calculation of the thermodynamic profiles resulting from introducing substituents that interacted with or displaced the ordered water showed that the gain of binding affinity was enthalpy driven. Detailed analysis of the energetics and binding site hydration networks revealed that the enthalpy change was governed by contributions that are commonly neglected in structure-based drug optimization. In particular, simulations suggested that displacement of water from a binding site to the bulk solvent can lead to large energy contributions. Our findings provide insights into the molecular driving forces of protein-ligand binding and strategies for rational drug design.

We present an atom-economic strategy to catalytically generate and intercept nitrile anion equivalents using hydrogen transfer catalysis. Addition of alpha,beta-unsaturated nitriles to a pincer-based Ru-H complex affords structurally characterized kappa-N-coordinated keteniminates by selective 1,4-hydride transfer. When generated insitu under catalytic hydrogenation conditions, electrophilic addition to the keteniminate was achieved using anhydrides to provide alpha-cyanoacetates in high yields. This work represents a new application of hydrogen transfer catalysis using alpha,beta-unsaturated nitriles for reductive C-C coupling reactions.

Here, a photocatalytic deoxygenative amidation protocol using readily available amine-boranes and carboxylic acids is described. Microglia-mediated neuroinflammation is one of the key mechanisms involved in acute brain injury and chronic neurodegeneration. This study investigated the inhibitory effects of 2-hydroxy-4-methylbenzoic anhydride (HMA), a novel synthetic derivative of HTB (3-hydroxy-4-trifluoromethylbenzoic acid) on neuroinflammation and underlying mechanisms in activated microglia in vitro and an in vivo mouse model of Parkinson’s disease (PD). In vitro studies revealed that HMA significantly inhibited lipopolysaccharide (LPS)-stimulated excessive release of nitric oxide (NO) in a concentration dependent manner. In addition, HMA significantly suppressed both inducible NO synthase and cyclooxygenase-2 (COX-2) at the mRNA and protein levels in LPS-stimulated BV-2 microglia cells. Moreover, HMA significantly inhibited the proinflammatory cytokines such as interleukin (IL)-1beta, IL-6, and tumor necrosis factor-alpha in LPS-stimulated BV-2 microglial cells. Furthermore, mechanistic studies ensured that the potent anti-neuroinflammatory effects of HMA (0.1, 1.0, and 10 mu M) were mediated by phosphorylation of nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha (I kappa B alpha) in LPS-stimulated BV-2 cells. In vivo evaluations revealed that intraperitoneal administration of potent neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 20 mg/kg, four times a 1 day) in mice resulted in activation of microglia in the brain in association with severe behavioral deficits as assessed using a pole test. However, prevention of microglial activation and attenuation of Parkinson’s disease (PD)-like behavioral changes was obtained by oral administration of HMA (30 mg/kg) for 14 days. Considering the overall results, our study showed that HMA exhibited strong anti-neuroinflammatory effects at lower concentrations than its parent compound. Further work is warranted in other animal and genetic models of PD for evaluating the efficacy of HMA to develop a potential therapeutic agent in the treatment of microglia-mediated neuroinflammatory disorders, including PD.

In this paper, solar binary chemical depolymerization, that is Solar Thermal Electrochemical Process (STEP), was implemented for an effective breaking of lignin into small molecules and hydrogen. Compared with the conventional unitary chemical thermolysis, solar binary chemical depolymerization of lignin has high efficiencies of the liquefaction and gasification with the low coke, and accompanied by the abundant production of hydrogen. And the reaction temperature of the STEP process was greatly lowered by an intervention of the electrolysis. The results showed that the total conversion and liquefaction of the lignin yielded 87.22% and 57.72% under a constant current of 0.4 A at 340 degrees C. Further characterizations show that lignin has been successfully decomposed into small molecules with high added-value and hydrogen by a combination of the thermolysis and electrolysis. And the particle size of aggregates and the color degree in the lignin aqueous solution was obviously decreased after the STEP process.

Selective modification of the hydroxyl groups of sugars has been a long-standing challenge due to their proximate relative reactivity. Herein, we report a TMSOTf-catalyzed selective acetylation of the non-anomeric hydroxyl groups of several per-O-TMS-protected sugar substrates while leaving their anomeric group unaffected. In addition to standing versatile by itself, the anomeric O-TMS group left intact can be functionalized to afford key sugar precursors such as imidate donors, which could otherwise be synthesized via a stepwise anomeric deprotection-functionalization procedure.

Oxidation of organic substrates is achieved in nature under mild conditions thanks to metalloenzymes but remains a challenge for chemists. Herein we show by UV-Vis spectroelectrochemistry that when (MnTPPCl)-T-III is electrochemically reduced to Mn-II in CH2Cl2 under O-2, a (MnO2)-O-II species is generated. Benzoic anhydride reacts with the latter triggering a catalytic current in cyclic voltammetry. Electrolysis on the catalytic wave in the presence of cyclooctene leads to its oxygenation or halogenation depending on the axial ligand present as reported here for the first time.

Well-functionalized monosaccharides were readily prepared, and highly efficient glycosylations using a common activator (NIS/ TfOH) were also presented.
