In this study, the novel compounds (2,5-dimethoxy-methanimine propyltriethoxysilane; S1 and 3-ethoxy-salicylaldimine propyltriethoxysilane; S2) derived from aminopropyltriethoxysilane (APTES) were synthesized by a simple and facile approach in order to modify mesoporous silica (MS) surface. Characterization of S1 and S2 was experimentally performed by FTIR, H-1-C-13 NMR spectroscopic methods and elemental analysis. The IR frequencies, H-1 and C-13 NMR chemical shift values for the optimized molecular geometries of S1 and S2 were calculated by using Density Functional Theory (DFT with B3LYP/6-311 + G (d, p) basis set), and the theoretical results were compared to the obtained experimental data. The experimental and theoretical results confirmed the proposed molecular structures of the synthesized S1 and S2 compounds. After that, the mesoporous silica material (MS) was synthesized according to the Stober procedure, and characterized by the elemental analysis, and the experimental and theoretical IR spectra. The surface of MS was successfully functionalized with S1 and S2 to produce more selective and effective adsorbent materials, and the structures of functionalized MSs (S1-MS and S2-MS) were investigated with FTIR spectroscopy and elemental analysis. The theoretical optimized molecular geometries, IR frequencies and H-1-C-13 NMR chemical shift values were calculated to execute the structural analysis of S1-MS and S2-MS. In addition, the calculations of the sum of electronic and thermal free energy (SETFE), dipole moment, the HOMO-LUMO energies, the molecular electrostatic potential (MEP), the solvent accessible surface (SAS) and atomic charge for the synthesized S1, S2, MS, S1-MS and S2-MS were accomplished by using DFT/B3LYP/6-311 + G (d, p) level of theory. Cyclin-dependent kinases play significant roles in cell cycle progression and are promising targets for cancer therapy. However, most potent CDK inhibitors lack the balance between efficacy and safety because of poor selectivity. Given the roles of CDK2 in tumorigenesis, selective CDK2 inhibition may provide therapeutic benefits against certain cancer. In this study, a series of 4-benzoylamino-1H-pyrazole-3-carboxamide derivatives were designed, synthesized, and evaluated. The most selective compound DC-K2in212 in this series exhibited high potency towards CDK2 and had effective anti-proliferative activity against A2058 melanoma cell line and MV4-11 leukemia cell line while exhibiting low toxic effect on human normal cell lines MRC5 and LX2. The molecular modeling illustrated that compound DC-K2in212 had the similar binding mode with CDK2 as C-73, the most selective CDK2 inhibitor reported so far, which might account for selectivity against CDK2 over CDK1. Further biological studies revealed that compound DC-K2in212 suppressed CDK2-associated downstream signaling pathway, blocked cell cycle progression, and induced cellular apoptosis. A series of tetrahydro-ss-carbolines substituted with an alkyl or acyl side chain was synthesized and screened for its antifungal activity against plant pathogenic fungi (Bipolaris oryzae, Curvularia lunata, Fusarium semitectum, and Fusarium fujikuroi). The structure activity relationship revealed that the substituent at the piperidine nitrogen plays an important role for increasing antifungal activities. In this series, 2-octyl-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole (3g) displayed potent antifungal activities with a minimum inhibitory concentration of 0.1 mu g/mL, including good inhibitory activity to the radial growth of fungus at a concentration of 100 mu g/mL compared to amphotericin B.

Solvent-free, atom-efficient, and mechanochemically activated reactions have emerged as synthetic strategy for sustainable chemistry. Herein we report a new mechanochemical approach for the amide coupling of carboxylic acids and amines, mediated by combination of (1-cyano-2-ethoxy-2-oxoethylidenaminooxy)-dimethylaminomorpholinocarbenium hexafluorophosphate (COMU) or N,N,N’,N’-tetramethylchloroformamidinium hexa-fluorophosphate (TCFH) and K2HPO4. The method delivers a range of amides in high yields (70-96%) and fast reaction rates. The reaction protocol is mild, keeps stereochemical integrity of the adjacent to carbonyl stereocenters, and streamlines isolation procedure for solid amide products. Minimal waste is generated due to the absence of bulk solvent. We show that K2HPO4 plays a dual role, acting as a base and a precursor of reactive acyl phosphate species. Amide bonds from hindered carboxylic acids and low-nucleophilic amines can be assembled within 90 minutes by using TCFH in combination with K2HPO4 or N-methylimidazole. The developed mechanochemical liquid-assisted amidation protocols were successfully applied to the challenging couplings of all six carboxylate functions of biotin[6]uril macrocycle with phenylalanine methyl ester, resulting in 80% yield of highly pure hexa-amide-biotin[6]uril. In addition, fast and high-yielding synthesis of peptides and versatile amide compounds can be performed in a safe and environmentally benign manner, as verified by green metrics.

Totally colorless, transparent, and highly refractive poly(phenylene thioether ether)s (PPTEs) containing thianthrene-2,7-disulfanyl moiety were developed in this study. A new 4,4′-diol aromatic compound, 2,7-(4,4′-diphenol)thiothianthrene (DPTT), with a high sulfur content was designed and synthesized to develop polymers with high refractive indices and high thermal stabilities. The PPTEs were prepared by polycondensation of DPTT with organic dihalogen compounds, such as 2,6-difluorobenzonitrile (DFBN) and 4,6-dichloro-2-(methylthio) pyrimidine (DCMP). The obtained polymers showed good thermal stabilities such as relatively high glass transition temperatures (T(g)s) in the range of 153-176 degrees C and 5% weight loss temperatures (T-5%) over 344 degrees C. The cut-off wavelengths (lambda(0)) of the PPTE films are shorter than 343 nm, and they have a transmittance higher than 93% at 550 nm. The combination of the pyrimidine and high sulfur-containing thianthrene derivatives provides the PPTEs with a high refractive index of 1.7204 and a small birefringence of 0.0106 at 637 nm.

Thirty-four seco-3’R,4’R-disubstituted-2′,2′-dimethyldihydropyrano[2,3-f]chromone (seco-DSP) derivatives were designed, synthesized and evaluated for chemo-reversal activity when combined with paclitaxel or vincristine in P-gp overexpressing A2780/T and KB-VIN drug-resistant cancer cell lines. Most of the compounds displayed moderate to significant MDR reversal activities. Compound 7e showed the most potent chemo-sensitization activity with more than 1471 reversal ratio at a concentration of 10 mu M, which was higher than verapamil (VRP) (212-fold). Unexpectedly the newly synthesized compounds did not show chemosensitization activities in a non-P-gp overexpressing cisplatin resistant human ovarian cancer cell line (A2780/CDDP), implying that the MDR reversal effects might be associated with P-gp overexpression. Moreover, the compounds did not exhibit significant anti-proliferative activities against non-tumorigenic cell lines (HUVEC, HOSEC and T29) compared to VRP at the tested concentration and might be safer than VRP. In preliminary pharmacological mechanism studies, the compounds increased accumulation of DOX and promoted P-gp ATPase activity in A2780/T cell lines. Western blot analysis indicated they did not affect the expression level of P-gp in the tested MDR cell lines. Synthesis of novel 4(3H)-quinazolinonyl aminopyrimidine derivatives has been achieved via quinazolinonyl enones which in turn were obtained from 2-acyl-4(3H)-quinazolinone. They have been assayed for biofilm inhibition against Gram-positive (methicillin-resistant Staphylococcus aureus (MRSA)) and Gram-negative bacteria (Acinetobacter baumannii). BACKGROUND: Chemical pesticides are the main measures for pest control, but have caused growing resistance of pests and brought a series of environmental problems. Development of high-efficient insecticidal molecules with novel scaffolds is therefore particularly urgent. RESULTS: Based on a [5 + 1] annulation reaction with 5-amino-1H-phenylpyrazole and dialkyl bromomalonate, 27 novel five-substituted 4,5-dihydropyrazolo[1,5-a]quinazolines were designed following the intermediate derivatization method and synthesized. Bioassay results indicated that most of the test compounds displayed good insecticidal activities against Plutella xylostella, Spodoptera frugiperda, and Solenopsis invicta. In particular, the insecticidal activities of compounds 4a, 4f, and 4m against P. xylostella [median lethal concentration (LC50) values ranged from 3.87 to 5.10 mg L-1] were comparable to that of indoxacarb (LC50 = 4.82 mg L-1). In addition, compounds 4a and 9e showed similar high insecticidal activities against Spodoptera frugiperda (mortality rate = 79.63% and 72.12%) at 100 mg L-1, comparable to that of fipronil (mortality rate: 68.44%); compound 9a showed possible delayed toxicity against Solenopsis invicta (mortality rate: 95.66%) after 5 days of treatment at 1.0 mg L-1. CONCLUSION: Due to their high insecticidal activities against P. xylostella, compound 4m, 4a, and 4f could be considered as qualified candidates for novel insecticide. Several other 4,5-dihydropyrazolo[1,5-a]quinazolines with relatively high bioactivity, such as compounds 9a and 9e, are also worth further optimization as potential insecticide or anticide candidates.

Several pyrrolo[2,3-b]pyridine-based B-RAF inhibitors are well known and some of them are currently FDA approved as anticancer agents. Based on the structure of these FDA approved B-V600E-RAF inhibitors, two series of pyrrolo[2,3-b]pyridine scaffold were designed and synthesized in attempt to develop new potent B-V600E-RAF inhibitors. The 38 synthesized compounds were biologically evaluated for their B-V600E-RAF inhibitory effect at single dose (10 mu M). Compounds with high percent inhibition were tested to determine their IC50 over (V600E)BRAF. Compounds 34e and 35 showed the highest inhibitory effect with IC50 values of 0.085 mu M and 0.080 mu M, respectively. Headed for excessive biological evaluation, the synthesized derivatives were tested over sixty diverse human cancer cell lines. Only compound 35 emerged as a potent cytotoxic agent against different panel of human cancer cell lines.
