Co-crystallizations can be utilized for generating new solid forms of a target substance in order to alter or enhance some specific bulk physical property. Generally, selection of the co-former (the necessary partner for the target molecule) is based on existing structural information about molecular recognition events involving complementary functional groups, and extensive experimental screening methods. In this study, we utilize structure-informatics in an attempt to predict if two different molecules will form a co-crystal or not. Our study is based on hydrogen-bond propensity (HBP), and the key premise of our approach rests on whether target-co-former interactions are more likely to take place than either target-target or co-formerco-former hydrogen bonds. We examined six different target molecules in combination with 25 possible co-formers each and used the HBP protocol for predicting if a co-crystal would form or not. The predictions were then compared with results from an experimental co-crystal screen of the 150 different combinations. The correct outcome was successfully predicted 92-95% of the time which shows that for this series of small molecules, HBP is a very reliable indicator for determining if a co-crystal will form between a target molecule and a particular co-former.

We report an efficient and straight forward access to nitrile substituted enol esters via ionic liquid [BMIM]OH mediated hydrocarboxylation of alkynylnitriles under mild conditions. This atom economical transition metal-free protocol gives an easy access to a variety of such enol esters with excellent regio and Z-stereoselectivity. Reusability of [BMIM]OH without losing of significant amount of yield is another noticeable feature of this article.

In the present work, the nanocrystalline particles of hydroxyapatite (HAp) using an easy alkoxide-based sol-gel technique including triethyl phosphate [PO (OC2H5)(3)] and Ca (NO3)(2)center dot 4H(2)O as P and Ca precursors have been synthesized. The sample characterization was performed by X-ray diffraction, Fourier transform-infrared analysis, scanning electron microscopy, thermal analysis (thermogravimetric analysis/differential thermal analysis), and elemental analysis of energy-dispersive X-ray analysis. It is interesting that single phase of HAp was obtained at a low firing temperature of 500 degrees C. Modified Scherrer equation as the Williamson-Hall method was applied for the measurement of crystallite size distributions and micro-strain of the sample. The determined crystallite size by complementary technique of transmission electron microscopy has good consistency with those obtained from the Scherrer formula. Moreover, we reported the one-pot synthesis of 3,4-disubstituted isoxazole-5(4H)-ones through the aqueous solution reaction of three components of ethyl acetoacetate, hydroxylamine hydrochloride and various aromatic aldehydes at room temperature. This protocol offers several advantages, including a simple work-up procedure, very short reaction times (under 25 min), in accordance with the principles of green chemistry, recyclability, excellent yields (87-98%) and environmentally friendly.

A series of novel alpha-aminophosphonate derivatives were synthesized via simple and an efficient method from the three-component condensation reaction of 5-aminoindan or 3,4-(methylenedioxy)aniline, aromatic aldehydes and diethyl phosphite by employing graphene nanosheets-silver nanoparticles (GNS-AgNPs) as catalyst under ultrasonication and solvent-free condition. GNS-AgNPs was prepared in situ by simultaneous reduction of graphene oxide (GO) and silver nitrate (AgNO3) using sodium borohydride (NaBH4) as a reducing agent. The surface characterization of GNS-AgNPs was done using X-ray diffraction (XRD), Fourier transform infrared (FTIR), UV-Vis spectroscopy (UV-Vis), Thermogravimetric analysis (TGA), Raman spectroscopy and Field emission scanning electron microscopy (FE-SEM). The GNS-AgNPs are recyclable up to fifth run with minimal loss of its activity. Convenient operational simplicity, mild conditions, excellent yields of products, consistent performance and the reusability of catalyst makes this protocol feasible and attractive.

Three-component reaction of methyl 4-aryl-2,4-dioxobutanoates with aromatic aldehyde and 2-aminoacetonitrile sulfate in glacial acetic acid in the presence of anhydrous sodium acetate afforded previously unknown (3-aroyl-2-aryl-4-hydroxy-5-oxo-2,5-dihydro-1H-pyrrol-1-yl)acetonitriles. The reaction involved intermediate formation of Schiff base, followed by Michael-type addition of the dioxo ester to the C=N bond and cyclization of the addition product, methyl 3-aroyl-4-aryl-4-(cyanomethylamino)-2-oxobutanoate. The cyclization product reacted with hydrazine hydrate at the aroyl carbonyl group to give the corresponding hydrazone which was converted in 1,4-dioxane to the cyclic form, [3,4-diaryl-6a-hydroxy-6-oxo-3a,4,6,6a-tetrahydropyrrolo[3,4-c]pyrazol-5(1H)-yl]acetonitrile, without elimination of the second water molecule. When the reaction of (3-aroyl-2-aryl-4-hydroxy-5-oxo-2,5-dihydro-1H-pyrrol-1-yl)acetomtriles with hydrazine hydrate was carried out in boiling acetic acid, [3,4-diaryl-6-oxo-2,6-dihydropyrrolo[3,4-c]pyrazol-5(4H)-yl]acetonitriles were obtained as a result of dehydration of initially formed [3,4-diaryl-6a-hydroxy-6-oxo-3a,4,6,6a-tetrahydropyrrolo[3,4-c]pyrazol-5(1H)-yl]acetonitriles as shown by special experiment. A facile three-component reaction of aromatic aldehyde, 2,2-dimethyl-1,3-dioxane-4,6-dione, and 6-amino-1,3-dimethyluracil was developed for the first time using beta-cyclodextrin (beta-CD) as a macrocyclic host for aldehyde and an efficient catalyst that leads to a batch of novel pyrano[2,3-d:6,5-d ‘]dipyrimidines (4a-k). The synthesis was accomplished with the aid of microwave irradiations in solvent-free conditions. The product obtained was in contrast to the previous report in which a similar reaction resulted in a mixture of benzylidenepyrimidine and bisaminopyrimidine analogs in the presence of triethylbenzylammonium chloride in an aqueous medium. The formation of the pyrano[2,3-d:6,5-d ‘]dipyrimidines may be in virtue of the property of beta-CD to construct new C-C and C-X (where X = heteroatom) bonds. Reusability of the catalyst up to three runs without any noteworthy change in catalytic activity is one of the main features of the reaction. Other noteworthy features are good to excellent yields, eco-friendly procedure, non-column chromatographic purification, and mild conditions.

New series of furan-thiazole hybrids (3a-f), thiazolo[2,3-c]-1,2,4-triazines (4a-f), their bioisosteres 1,3,4-thiadiazolo[2,3-c]-1,2,4-triazines (8a-d) and 1,2,4-triazino[4,3-b]-1,2,4-triazines (13a-e) were designed, synthesized and evaluated for their in vitro antitumor activities at the National Cancer Institute (NCI, USA). Among the synthesized compounds, 3d was found to exhibit promising broad spectrum antitumor activity (GI(50) MG-MID = 14.22 mu M) in a five-dose assay against the full panel NCI-cancer cell lines. 3d displayed higher antitumor activity against most tested cancer cell lines than 5-FU as reference. COMPARE analysis and molecular electrostatic potential computational study revealed that 3d probably exerts its antitumor properties through DNA binding similar to Clomesone. Further DNA binding studies using fluorescent terbium (Tb+3) probe revealed increased fluroresence of DNA-3d-Tb+3 mixture due to damage of the double-stranded DNA. Also, UV-vis absorption study was conducted which showed hyperchromic shift in DNA absorption confirming 3d-induced DNA damage. The assessed potency of 3d-induced DNA damage of calf thymus DNA showed a concentration as low as 2.04 ng/mL for a detectable DNA damage. Moreover, in silico calculation of physicochemical properties and druglikeness were in compliance to Lipinski’s rule.

Solvent-free, atom-efficient, and mechanochemically activated reactions have emerged as synthetic strategy for sustainable chemistry. Herein we report a new mechanochemical approach for the amide coupling of carboxylic acids and amines, mediated by combination of (1-cyano-2-ethoxy-2-oxoethylidenaminooxy)-dimethylaminomorpholinocarbenium hexafluorophosphate (COMU) or N,N,N’,N’-tetramethylchloroformamidinium hexa-fluorophosphate (TCFH) and K2HPO4. The method delivers a range of amides in high yields (70-96%) and fast reaction rates. The reaction protocol is mild, keeps stereochemical integrity of the adjacent to carbonyl stereocenters, and streamlines isolation procedure for solid amide products. Minimal waste is generated due to the absence of bulk solvent. We show that K2HPO4 plays a dual role, acting as a base and a precursor of reactive acyl phosphate species. Amide bonds from hindered carboxylic acids and low-nucleophilic amines can be assembled within 90 minutes by using TCFH in combination with K2HPO4 or N-methylimidazole. The developed mechanochemical liquid-assisted amidation protocols were successfully applied to the challenging couplings of all six carboxylate functions of biotin[6]uril macrocycle with phenylalanine methyl ester, resulting in 80% yield of highly pure hexa-amide-biotin[6]uril. In addition, fast and high-yielding synthesis of peptides and versatile amide compounds can be performed in a safe and environmentally benign manner, as verified by green metrics.

In this article, we report an efficient and mild synthetic route for the construction of substituted quinazolines from functionalized 2-aminobenzophenones with various benzaldehydes usingcat. TMSOTf and hexamethyldisilazane (HMDS) under neat, metal-free and microwave irradiation conditions in which gaseous ammonia was formedin situ. This synthetic protocol provided the desired quinazolines with a broad substrate scope in good to excellent yields. Some structures were confirmed by X-ray single-crystal diffraction analysis.

Several pyrrolo[2,3-b]pyridine-based B-RAF inhibitors are well known and some of them are currently FDA approved as anticancer agents. Based on the structure of these FDA approved B-V600E-RAF inhibitors, two series of pyrrolo[2,3-b]pyridine scaffold were designed and synthesized in attempt to develop new potent B-V600E-RAF inhibitors. The 38 synthesized compounds were biologically evaluated for their B-V600E-RAF inhibitory effect at single dose (10 mu M). Compounds with high percent inhibition were tested to determine their IC50 over (V600E)BRAF. Compounds 34e and 35 showed the highest inhibitory effect with IC50 values of 0.085 mu M and 0.080 mu M, respectively. Headed for excessive biological evaluation, the synthesized derivatives were tested over sixty diverse human cancer cell lines. Only compound 35 emerged as a potent cytotoxic agent against different panel of human cancer cell lines.
