Totally colorless, transparent, and highly refractive poly(phenylene thioether ether)s (PPTEs) containing thianthrene-2,7-disulfanyl moiety were developed in this study. A new 4,4′-diol aromatic compound, 2,7-(4,4′-diphenol)thiothianthrene (DPTT), with a high sulfur content was designed and synthesized to develop polymers with high refractive indices and high thermal stabilities. The PPTEs were prepared by polycondensation of DPTT with organic dihalogen compounds, such as 2,6-difluorobenzonitrile (DFBN) and 4,6-dichloro-2-(methylthio) pyrimidine (DCMP). The obtained polymers showed good thermal stabilities such as relatively high glass transition temperatures (T(g)s) in the range of 153-176 degrees C and 5% weight loss temperatures (T-5%) over 344 degrees C. The cut-off wavelengths (lambda(0)) of the PPTE films are shorter than 343 nm, and they have a transmittance higher than 93% at 550 nm. The combination of the pyrimidine and high sulfur-containing thianthrene derivatives provides the PPTEs with a high refractive index of 1.7204 and a small birefringence of 0.0106 at 637 nm.

In subsurface bioremediation, electron donor addition promotes microbial Fe(lII)-oxide mineral reduction that could change soil pore structure, release colloids, and alter soil surface properties. These processes in turn may impact bioremediation rates and the ultimate fate of contaminants. Columns packed with water-stable, Fe-oxide-rich soil aggregates were infused with acetate-containing artificial groundwater and operated for 20 d or 60 d inside an anoxic chamber. Soluble Fe(II) and soil colloids were detected in the effluent within one week after initiation of the acetate addition, demonstrating Fe(III)-bioreduction and colloid formation. Diffusible Br-, less diffusible 2,6-difluorobenzoate (DFBA), and non-diffusible silica-shelled silver nanoparticles (SSSNP) were used as tracers in transport experiments before and after the bioreduction. The transport of Br- was not influenced by the bioreduction. DFBA showed earlier breakthrough and less tailing after the bioreduction, suggesting alterations in flow paths and soil surface chemistry during the 20-d bioreduction treatment. Similarly, the bioreduction increased the transport of SSSNP very significantly, with mass recovery increasing from 1.7% to 25.1%. Unexpectedly, the SSSNP was completely retained in the columns when the acetate injection was extended from 20 to 60 d, while the mass recovery of DFBA decreased from 89.1% to 84.1% and Br showed no change. The large change in the transport of SSSNP was attributed to soil aggregate breakdown and colloid release (causing mechanical straining of SSSNP) and the exposure of iron oxide surfaces previously unavailable within aggregate interiors (facilitating attachment of SSSNP). These results suggest a time dependent fashion of microbial effect on the transport of diffusivity-varying tracers. (C) 2018 Elsevier Ltd. All rights reserved.

The 1,3,4-thiadiazole core has been mainly used as a pharmacological scaffold in medicinal chemistry. A series of Schiff bases derived from 5-substituted-1,3,4-thiadiazole-2-amine were designed and synthesized to investigate their biological activities. Structures of compounds were clarified with FTIR, H-1 NMR and elemental analysis. Due to the importance of this core in pharmacology, all these newly synthesized compounds were tested for different biological properties at the same time. Compound 3A ((E)-N-(2,5-dimethoxybenzylidene)-5-(4-methoxyquinolin-2-yl)-1,3,4-thiadiazol-2-amine) and compound 4A ((E)-N-(2,5-dimethoxybenzylidene)-5-(3-methylbenzofuran-2-yl)-1,3,4-thiadiazol-2-amine) possessed high DNA protective ability against oxidative Fenton mixture. Compound 1A ((E)-N-(2,5-dimethoxybenzylidene)-5-(benzo[b]thiophen-2-yl)-1,3,4-thiadiazol-2-amine) and compound 2B ((E)-2-((5-(1H-indol-2-yl)-1,3,4-thiadiazol-2-ylimino)methyl)-6-methoxyphenol) showed strong antimicrobial activity against S. epidermidis. The most effective compound was detected as compound 3A which exhibited cytotoxicity on both PC-3 and MDA-MB-231 cancer cell lines. The IC50 of this compound was calculated as 370.7 mu M and 505.1 mu M for MDA-MB-231 and PC-3 cells, respectively. Molecular docking studies were also performed to examine the understanding of the mechanism behind the anti-cancer and anti-bacterial properties. For further study, compound 3A has the potential for utilization with chemotherapy drugs to establish a more efficient therapy strategy with minimum cytotoxicity against cancer cells. A series of novel dihydropyranoindole derivatives containing sulphonamide group were designed, synthesized and evaluated for in-vitro anti-cholinesterase activity. The result showed that all the compounds exhibited potent acetylcholinesterase (AChE) activity (IC50 = 0.41-8.79 mu M) while demonstrated moderate to good activity for butyrylcholinesterase (BuChE) (IC50 = 1.17-30.17 mu M). The tested compounds exhibited selectivity towards AChE over BuChE. Compound 5o was most potent towards both AChE (IC50 = 0.41 mu M) and BuChE (IC50 = 1.17 mu M) when compared to standard galantamine and rivastigmine. Enzyme kinetics and molecular docking studies revealed that compound 5o shows mixed type inhibition and binds to peripheral anionic site (PAS) and the catalytic sites (CAS) of both the enzymes. Furthermore, cell viability studies were also performed against N2a cells along with neuroprotection studies against H2O2 in the same cell line. Antioxidant studies using DPPH radical and H2O2 were also performed which revealed that all compounds possessed some antioxidant activity. Also, DNA damage protection assay for compound 5o was performed implying that compound 5o was protective in nature. ADME studies were also performed which demonstrated good pharmacokinetics. These findings indicated that dihydropyranoindole derivatives could be possible drug lead in the search for new multifunctional AD drugs.

Substituted 1,2,4-triazole nucleus is common in several drugs used in a variety of clinical conditions including infections, hypoglycemia, hypertension and cancer. In this study, we synthesized 1,2,4-triazole and its 16 hydrazone derivatives (B1-B16), characterized them by IR, NMR and Mass spectroscopy, and evaluated their radical scavenging and anti-inflammatory activities in vitro and in vivo. Out of 16 derivatives, five (B1, B5, B6, B9, and B13) demonstrated a significant radical scavenging and anti-inflammatory activity in vitro. B6, which possessed two electron-donating hydroxyl groups, was most active among all. Molecular docking and MD simulation of the complex of B6 with prostaglandin-endoperoxide synthase (PTGS) or cyclooxygenase (COX) showed that B6 occupied celecoxib binding site in COX with high affinity (the binding free energy of the complex with COX-1 was -10.5, and -11.2 kcal/mol with COX-2). Maximum anti-inflammatory activity was also shown by the B6 derivative in vivo, in the rat model of carrageenan-induced inflammation. B6, along with four other derivatives (B1, B5, B9 and B13) exhibited 80-90% free radical scavenging activity. The IC50 values of these compounds were >= 40 mu M. Griess nitrite and dichloro-dihydro-fluorescein-diacetate assays suggested a significant inhibition of nitric oxide and reactive oxygen species, especially by B6 and B9. Taken together, out of 16 derivatives, B6 is reported to have highest anti-inflammatory and antioxidant activity at a low dose level, which may be attributed to its two electron-donating hydroxyls. B6 is proposed to be an important scaffold for the synthesis of new drugs against PTGS for use in a myriad of inflammatory and infectious diseases. Communicated by Ramaswamy H. Sarma

This study presents an efficient synthesis of 3-[6-(substituted-phenyl)-[1,2,4]triazolo[3,4-b][1,3,4] thiadiazol-3-yl]-1H-indazole via dehydrative condensation with cyclization of 4-amino-5-(1H-indazol-3-yl)-4H-[1,2,4]triazole-3-thiol and fluorinated or nonfluorinated carboxylic acids in presence of phosphorous oxychloride. The multistep reaction pathway proceeds through different compounds. Present synthesis has the advantages of easily accessible starting materials, convenient synthesis, simple reaction condition, wider substrate scope, and higher yield (75% to 90% isolated).

The cross-coupling of sodium (hetero)aryl carboxylates with (hetero)aryl chlorides proceeds with 1 mol % palladium catalyst and does not require inorganic base, silver salts, or copper salts. This coupling uses two low energy partners, and the only stoichiometric byproducts are carbon dioxide and sodium chloride. The substrate scope includes less activated aryl chlorides and carboxylates (>25 examples). The palladium loading could be reduced to 0.1 mol %, and Buchwald-style precatalysts could be used.
