High-mannose (Man(9)GlcNAc(2)) is the main carbohydrate unit present in viral envelope glycoproteins such as gp120 of HIV and the GP1 of Ebola virus. This oligosaccharide comprises the Man(9)epitope conjugated to two terminalN-acetylglucosamines by otherwise rarely-encountered beta-mannose glycosidic bond. Formation of this challenging linkage is the bottleneck of the few synthetic approaches described to prepare high mannose. Herein, we report the synthesis of the Man(9)epitope with both alpha and beta configurations at the reducing end, and subsequent evaluation of the impact of this configuration on binding to natural receptor of high-mannose, DC-SIGN. Using fluorescence polarization assays, we demonstrate that both anomers bind to DC-SIGN with comparable affinity. These relevant results therefore indicate that the more synthetically-accesible Man(9)alpha epitope may be deployed as ligand for DC-SIGN in bothin vitroandin vivobiological assays.

We described the chemical synthesis of a sulfated trisaccharide repeating unit of fucosylated chondroitin sulfate (FCS), which has significant anticoagulant activity. Well-functionalized monosaccharides were readily prepared, and highly efficient glycosylations using a common activator (NIS/ TfOH) were also presented. The synthesized trisaccharide 4 could be used to extend oligosaccharide sequences.

Trace volatile organic compounds (VOCs) such as aldehydes and aromatics significantly deteriorate the indoor air quality especially in newly-decorated buildings. The universal water vapor would greatly decrease the adsorption capacity of ad-sorbents for trace VOCs via competitive adsorption and capillary condensation. Herein, we developed a strategy to subsequently modify the internal and external surface of CAU-1, an aluminum-based metal-organic framework (MOF), to significantly improve its humidity resistance. First, anhydrides were used to transfer amino groups of CAU-1 into hydrophobic groups via amide reaction. Second, polydimethylsiloxane was used to form a hydorphobic coating on its external surface. The adsorption capacity of as-modified CAU-1 for 1 ppm toluene increased about 6 times under 50% relative humidity (RH) at 298 K, even much higher than those of common adsorbents including hydrophobic ZSM-5. In addition, it could be completely regenerated below 373 K, about 65 K lower than that required for regenerating ZSM-5 under the same condition, which not only greatly reduces the energy consumption but also improves the safety and feasibility of in-situ regeneration of adsorbents in indoor environment.

Axially chiral phenols are attractive targets in organic synthesis. This motif is central to many natural products and widely used as precursors to, or directly, as chiral ligands and catalysts. Despite their utility few simple catalytic methods are available for their synthesis in high enantiopurity. Herein the atropselective acylation of a range of symmetric biaryl diols is investigated using isothiourea catalysis. Studies on a model biaryl diol substrate shows that the high product er observed in the process is a result of two successive enantioselective reactions consisting of an initial enantioselective desymmetrization coupled with a second chiroablative kinetic resolution. Extension of this process to a range of substrates, including a challenging tetraorthosubstituted biaryl diol, led to highly enantioenriched products (14 examples, up to 98:2 er), with either HyperB(TM) or B-TM identified as the optimal catalyst depending upon the substitution pattern within the substrate. Computation has been used to understand the factors that lead to high enantiocontrol in this process, with maintenance of planarity to maximize a 1,5-S…O interaction within the key acyl ammonium intermediate identified as the major feature that determines atropselective acylation and thus product enantioselectivity.

Axially chiral atropisomeric compounds are widely applied in asymmetric catalysis and medicinal chemistry, and efficient methods for their synthesis are in high demand. This applies in particular to atropisomers derived from five-membered aromatic rings because their lower barrier for rotation among the biaryl axis limits their asymmetric synthesis. We report here an enantioselective C-H functionalization method using our chiral RhJasCp complex for the synthesis of the biaryl atropisomer types that can be accessed from three different five-membered-ring heterocycles.

The microporous framework copper silicates CuSH-1NaK (1) and SGU-29 (2) – prepared using hydrothermal conditions – are effective heterogeneous oxidation catalysts for producing benzaldehyde (BzAld) and benzoic acid (BzAcid) from benzyl alcohol (BzOH), under mild reaction conditions (approximately atmospheric pressure, 70 degrees C). The copper silicate heterogeneous catalysts triggered a radical reaction mechanism leading to the target products; in particular, BzAcid was formed in up to 94 % yield and 100 % selectivity from BzAld. An electrochemical synthesis method for the selective N1-acylation of indazoles has been developed. This anion pool approach electrochemically reduces indazole molecules generating indazole anions and H-2. Acid anhydrides are then introduced to the solution resulting in selective acylation of the N1-position of the indazoles. This procedure can also be applied to the acylation of benzimidazoles and indoles. The reaction can also be performed using a 9 V battery without loss of reaction efficiency.

Oxidation of organic substrates is achieved in nature under mild conditions thanks to metalloenzymes but remains a challenge for chemists. Herein we show by UV-Vis spectroelectrochemistry that when (MnTPPCl)-T-III is electrochemically reduced to Mn-II in CH2Cl2 under O-2, a (MnO2)-O-II species is generated. Benzoic anhydride reacts with the latter triggering a catalytic current in cyclic voltammetry. Electrolysis on the catalytic wave in the presence of cyclooctene leads to its oxygenation or halogenation depending on the axial ligand present as reported here for the first time.
