Two catalytic asymmetric vinylogous aldol-type reactions of aldehydes with ally! phosphonate and allyl sulfone have been uncovered in good to high yields for the first time, The bulky ligand-(R)-DTBM-SEGPHOS-was found to be the key to perfectly control both regio- and enantioselectivities. Transformations of the vinylogous products (including Horner-Wadsworth-Emmons and Julia olefinations) were successfully realized by virtue of the phosphonate and sulfone moieties. Nano copper ferrite catalyst is prepared and characterized by scanning electron microscopy, energy dispersive X-ray, X-ray diffraction, vibrational sample magnetometry, and Fourier transform infrared. The catalytic activity is probed for cross-dehydrogenative coupling of aromatic aldehydes in the presence of tert-butyl hydroperoxide as the oxidant. This catalytic protocol appears as a simple, rather cheap, clean, and efficient practical strategy for the synthesis of symmetrical anhydrides, with proper efficiency (66%). Arenes with four different contiguous substituents, i.e. 1,2,3,4-tetrasubstituted arenes, are commonly found in bioactive compounds, but they are nontrivial to access via conventional methods. Through addressing the meta constraint in the palladium/norbornene (Pd/NBE) cooperative catalysis, which is the difficulty of tolerating a sizable meta substituent in aryl halide substrates, here a modular and regioselective approach is realized for preparing 1,2,3,4-tetrasubstituted arenes. One key is the use of a C2-amide-substituted NBE, and a combined experimental and computational study reveals its role in promoting the NBE insertion and the ortho C-H metalation steps. The scope is broad: a variety of electrophiles and nucleophiles could be introduced to the ortho and ipso positions, respectively, with 1,4-disubstituted aryl halides, leading to diverse unsymmetrical contiguous tetrasubstituted arenes. Application of this approach has been demonstrated in streamlined syntheses of several bioactive compounds.

Infectious diseases due to multidrug-resistant pathogens, particularly carbapenem-resistant Enterobacteriaceae (CREs), present a major and growing threat to human health and society, providing an urgent need for the development of improved potent antibiotics for their treatment. We describe the design and development of a new class of aminoglycoside antibiotics culminating in the discovery of propylamycin. Propylamycin is a 4′-deoxy-4′-alkyl paromomycin whose alkyl substituent conveys excellent activity against a broad spectrum of ESKAPE pathogens and other Gram-negative infections, including CREs, in the presence of numerous common resistance determinants, be they aminoglycoside modifying enzymes or rRNA methyl transferases. Importantly, propylamycin is demonstrated not to be susceptible to the action of the ArmA resistance determinant whose presence severely compromises the action of plazomicin and all other 4,6-disubstituted 2-deoxystreptamine aminoglycosides. The lack of susceptibility to ArmA, which is frequently encoded on the same plasmid as carbapenemase genes, ensures that propylamycin will not suffer from problems of cross-resistance when used in combination with carbapenems. Cell-free translation assays, quantitative ribosome footprinting, and X-ray crystallography support a model in which propylamycin functions by interference with bacterial protein synthesis. Cell-free translation assays with humanized bacterial ribosomes were used to optimize the selectivity of propylamycin, resulting in reduced ototoxicity in guinea pigs. In mouse thigh and septicemia models of Escherichia coli, propylamycin shows excellent efficacy, which is better than paromomycin. Overall, a simple novel deoxy alkyl modification of a readily available aminoglycoside antibiotic increases the inherent antibacterial activity, effectively combats multiple mechanisms of aminoglycoside resistance, and minimizes one of the major side effects of aminoglycoside therapy.

The catalytic efficiency of zeolitic imidazolate framework (ZIF-8) has been explored by making it multifunctional. Dual active sites were incorporated such as acid (dodecatungstophosphoric acid, DTP) and metal (Pd degrees) to prepare 5% Pd-DTP@ZIF-8. DTP was encapsulated inside the cage of ZIF-8 by in-situ and bottle around the ship approach whereas Pd was loaded ex-situ by simple wet impregnation method. The catalytic efficiency was tested for one pot synthesis of 3-phenyl propyl benzoate (3-PPB), a perfumery compound, from cinnamyl alcohol and benzoic anhydride. Trans-esterification of cinnamyl alcohol with benzoic anhydride gives cinnamyl benzoate which on further hydrogenation gives 3-PPB. Three different supports were screened such as ZIF-8, K10 and MCF out of which ZIF-8 showed the maximum activity because of its high surface area and smaller pore diameter. Further Pd, Ni and Cu metals were studied for selective hydrogenation of C=C bond among which 5% Pd-DTP@ZIF-8 gave almost 98% conversion of cinnamyl benzoate to 3-PPB with 93% selectivity. Fresh and spent catalysts were characterized by various techniques. 5% Pd-DTP@ZIF-8 showed anti-leaching property with great thermal stability. The turn over frequency and turn over number of the catalyst was observed to be 0.058 s(-1) and 835, respectively. A kinetic model was developed with good fit using LHHW mechanism and the activation energy calculated as 17.45 kcal/mol for hydrogenation step. Thus, the reaction was found to be kinetically controlled. The entire process is green.

H-beta Zeolites, doped with various metal salts, were investigated as heterogeneous catalysts for continuous-flow Friedel-Crafts acylation reactions. It was found that Zr-beta zeolites possessed excellent catalyst activity and stability, which enabled its use as a catalyst, up to 5 days, for Friedel-Crafts reactions between various activated arenes and acid anhydrides. Self-assembling peptides and oligonucleotides have given rise to synthetic materials with several applications in nanotechnology. Aggregation of synthetic oligosaccharides into well-defined architectures has not been reported even though natural polysaccharides, such as cellulose and chitin, are key structural components of biomaterials. Here, we report that six synthetic oligosaccharides, ranging from dimers to hexamers, self-assemble into nanostructures of varying morphologies and emit within the visible spectrum in an excitation-dependent manner. Well-defined differences in chain length, monomer modification, and aggregation methods yield glycomaterials with distinct shapes and properties. The excitation-dependent fluorescence in a broad range within the visible spectrum illustrates their potential for use in optical devices and imaging applications. We anticipate that our systematic approach of studying well-defined synthetic oligosaccharides will form the foundation of our understanding of carbohydrate interactions in nature.

This contribution describes sulfoximine (SFI) derivatives as a novel type of acceptor for efficient thermally activated delayed fluorescence (TADF) emitters. SFI presents a unique advantage by facile introduction of versatile functionalities on its secondary amine, which allows delicate manipulation of TADF properties. In a proof of principle study, three green/yellowish green TADF emitters, namely DMAC-SFIP, DMAC-SFIC and DMAC-SFIS were developed with different electron-withdrawing N-substituents based on simple donor-acceptor (D-A) typed structure. A look into the molecular conformations indicates the existence of different types of weak intramolecular interactions to suppress nonradiative decay. More importantly, effective mixing between charge transfer (CT) and locally excited (LE) triplet states is observed for DMAC-SFIP, DMAC-SFIC due to suitable electron-deficiency in the SFI segment, resulting in acceleration of the reverse intersystem crossing (RISC) rate. Consequently, these new TADF emitters render excellent organic light-emitting diode (OLED) performances with external quantum efficiencies (EQEs) up to 20.4% and small efficiency roll-off.

We herein report the development of a conformationally defined, electron-rich, C-2-symmetric, P-chiral bisphosphorus ligand, ArcPhos, by taking advantage of stereoelectronic effects in ligand design. With the Rh-ArcPhos catalyst, excellent enantioselectivities and unprecedentedly high turnovers (TON up to 10 000) were achieved in the asymmetric hydrogenation of aliphatic carbocyclic and heterocyclic tetrasubstituted enamides, to generate a series of chiral cis-2-alkyl-substituted carbocyclic and heterocyclic amine derivatives in excellent enantiomeric ratios. This method also enabled an efficient and practical synthesis of the Janus kinase inhibitor (R)-tofacitinib.
