Organosilicon compounds have been extensively utilized both in industry and academia. Studies on the syntheses of diverse organosilanes is highly appealing. Through-space metal/hydrogen shifts allow functionalization of C-H bonds at a remote site, which are otherwise difficult to achieve. However, until now, an aryl to alkyl 1,5-palladium migration process seems to have not been presented. Reported herein is the remote olefination, arylation, and borylation of a methyl group on silicon to access diverse vinyl-, benzyl-, and borylsilanes, constituting a unique C(sp(3))-H transformation based on a 1,5-palladium migration process.

Axially chiral phenols are attractive targets in organic synthesis. This motif is central to many natural products and widely used as precursors to, or directly, as chiral ligands and catalysts. Despite their utility few simple catalytic methods are available for their synthesis in high enantiopurity. Herein the atropselective acylation of a range of symmetric biaryl diols is investigated using isothiourea catalysis. Studies on a model biaryl diol substrate shows that the high product er observed in the process is a result of two successive enantioselective reactions consisting of an initial enantioselective desymmetrization coupled with a second chiroablative kinetic resolution. Extension of this process to a range of substrates, including a challenging tetraorthosubstituted biaryl diol, led to highly enantioenriched products (14 examples, up to 98:2 er), with either HyperB(TM) or B-TM identified as the optimal catalyst depending upon the substitution pattern within the substrate. Computation has been used to understand the factors that lead to high enantiocontrol in this process, with maintenance of planarity to maximize a 1,5-S…O interaction within the key acyl ammonium intermediate identified as the major feature that determines atropselective acylation and thus product enantioselectivity.

Non-enzymatic acylative kinetic resolution of racemic 2,2-dimethyl-substituted nitroaldol (Henry) adducts was achieved using cyclohexanecarboxylic anhydride in the presence of chiral guanidine catalyst. The systematic evaluation of the substituents on the aromatic rings of the substrates were conducted to clarify the influence of the substituents. The reaction was smoothly proceeded with moderate to good s-values, regardless of the kinds of the substituents. Based on the absolute configuration of the resulted product, the transition state of the reaction was predicted, and it was important the interaction between the nitro group on the substrate and the cation face of the acyl-guanidium intermediate to determine the direction of the substrate.

The microporous framework copper silicates CuSH-1NaK (1) and SGU-29 (2) – prepared using hydrothermal conditions – are effective heterogeneous oxidation catalysts for producing benzaldehyde (BzAld) and benzoic acid (BzAcid) from benzyl alcohol (BzOH), under mild reaction conditions (approximately atmospheric pressure, 70 degrees C). The copper silicate heterogeneous catalysts triggered a radical reaction mechanism leading to the target products; in particular, BzAcid was formed in up to 94 % yield and 100 % selectivity from BzAld. The type of oxidant and solvent influenced the catalytic performances.

This work entails deep red emitting Eu-III based complexes with a variety of ternary beta-diketonate ligands and 1,10-phenanthroline as the ancillary ligand in the system. The solid state structure and spectroscopic analysis has been outlaid in terms of photoluminescence and crystallography. A luminescence quantum efficiency of 50% was obtained for the [tris-(4,4,4-trifluoro-1-chlorophenyl-butanedione)mono-(1,10-phenanthroline)europium(iii)] complexes. Moreover, complexes [tris-(2,2,6,6-tetramethyl-heptanedione)mono-(1,10-phenanthroline) europium(iii)] and {[hexa-(benzyl carboxylic acid) bis-(1,10-phenanthroline)di-europium(iii)]-mu-[kappa(2)-O,O ‘-(benzyl carboxylic acid)](2)} were found to also have quantum yields of 9% and 28% with respective sensitization efficiencies of 85%, 15% and 58%. These results were articulated with crystallographic details pertaining to the nature of coordination and the effect of steric and electronic properties thereof which somewhat impacts the Eu-N bond distances. A symmetry correlation was drawn between the crystallographic data and the photoluminescence data.

A bifunctional amine/squaramide catalyst promoted direct aldol addition of an hydantoin surrogate to pyridine 2-carbaldehyde N-oxides to afford adducts bearing two vicinal tertiary/quaternary carbons in high diastereo- and enantioselectivity (d.r. up to >20:1; ee up to 98 %) is reported. Acid hydrolysis of adducts followed by reduction of the N-oxide group yields enantiopure carbinol-tethered quaternary hydantoin-azaarene conjugates with densely functionalized skeletons. DFT studies of the potential energy surface (B3LYP/6-31+G(d)+CPCM (dichloromethane)) of the reaction correlate the activity of different catalysts and support an intramolecular hydrogen-bond-assisted activation of the squaramide moiety in the transition state of the catalytic reaction.

New RNA modifications are needed to advance our toolbox for targeted manipulation of RNA. In particular, the development of high-performance reporter groups facilitating spectroscopic analysis of RNA structure and dynamics, and of RNA-ligand interactions has attracted considerable interest. To this end, fluorine labeling in conjunction with F-19-NMR spectroscopy has emerged as a powerful strategy. Appropriate probes for RNA previously focused on single fluorine atoms attached to the 5-position of pyrimidine nucleobases or at the ribose 2 ‘-position. To increase NMR sensitivity, trifluoromethyl labeling approaches have been developed, with the ribose 2 ‘-SCF3 modification being the most prominent one. A major drawback of the 2 ‘-SCF3 group, however, is its strong impact on RNA base pairing stability. Interestingly, RNA containing the structurally related 2 ‘-OCF3 modification has not yet been reported. Therefore, we set out to overcome the synthetic challenges toward 2 ‘-OCF3 labeled RNA and to investigate the impact of this modification. We present the syntheses of 2 ‘-OCF3 adenosine and cytidine phosphoramidites and their incorporation into oligoribonucleotides by solid-phase synthesis. Importantly, it turns out that the 2 ‘-OCF3 group has only a slight destabilizing effect when located in double helical regions which is consistent with the preferential C3 ‘-endo conformation of the 2 ‘-OCF3 ribose as reflected in the (3)J (H1 ‘-H2 ‘) coupling constants. Furthermore, we demonstrate the exceptionally high sensitivity of the new label in F-19-NMR analysis of RNA structure equilibria and of RNA-small molecule interactions. The study is complemented by a crystal structure at 0.9 angstrom resolution of a 27 nt hairpin RNA containing a single 2 ‘-OCF3 group that well integrates into the minor groove. The new label carries high potential to outcompete currently applied fluorine labels for nucleic acid NMR spectroscopy because of its significantly advanced performance.
