Platelet thrombosis is the main pathogeny resulting in the low curability of ischemic stroke, a leading cause of mortality and disability worldwide. Metformin, a biguanide derivative that is the first-line oral medicine for type 2 diabetes, alleviates the severity of ischemic stroke in diabetic patients and suppresses platelet activation in experimental animal model. However, the clinical implementation of commercial biguanide analogs for stroke related to platelet thrombosis remains challenging due to its weak potency, poor pharmacokinetic characteristics and possible hypoglycemia. Here, twenty-three biguanide derivatives were designed and synthesized based on the principles of bioisosteres. These derivatives were evaluated for the activity of antiplatelet thrombosis in vivo. We found that N-tri-fluoromethanesulfonyl biguanide derivative, compound b10, uniquely prevented cerebral infarction as well as neuronal function injury, and significantly decrease the mortality rate of ischemic stroke in the middle cerebral artery occlusion mice without significant side effects. We verified that b10 directly inhibited platelets thrombus formation and decreased the compactness of stroke thrombi. Particularly, b10 exhibited good potency to inhibit human platelet activation including platelet aggregation, adhesion, pseudopodia formation, integrin GPIIb/IIIa activation, CD62P expression and clot retraction. Meanwhile, the pharmacokinetics assessment showed that b10 had satisfying pharmacological characteristics including a longer duration and a higher oral absorption ratio than its parent compound. In addition, b10 remarkably ameliorated not only stroke related to platelet thrombosis but also carotid artery thrombus formation. It is concluded that the novel potent antiplatelet thrombotic agent derived from biguanide is a promising candidate for stroke treatment. (C) 2020 Elsevier Masson SAS. All rights reserved.

Because 6-amino-6-deoxy amylose is an important intermediate for many materials, developing a practical pathway to easily prepare it in bulk is of significance. The synthesis of 6-amino-6-deoxy amylose included three reactions. In the chlorination reaction, the crude product could be fully de-esterified under the improved conditions, and the reaction mechanism was re-understood. The azido group was reduced with sodium borohydride in which the reaction conditions were studied in detail. With the improved synthetic pathway, 6-amino-6-deoxy amylose could be easily prepared. The 6-amino-6-deoxy amylose was characterized via its benzamide derivative with(1)H NMR,H-1-C-13 HSQC NMR, and(1)H-H-1 COSY NMR, confirming the hydroxyl group at C(6)of amylose was converted to amino group with a degree of substitution of 98.2%. Additionally, the 6-amino-6-deoxy amylose was analyzed toward iodine reagent and X-ray diffraction, and the corresponding results showed that it was different from amylose in suprastructure and crystal structure.

A protocol of highly regio- and enantioselective copper-catalyzed hydroacylation of the non-terminal CC bond in 1,1-disubstituted terminal allenes with anhydrides has been developed. Both aromatic and aliphatic carboxylic anhydrides are applicable to the efficient construction of all carbon quarternary centers connected with a versatile C-C bond and a useful ketone functionality. Polyfunctional arenes are an important part of the chemical value chain. To improve the efficiency of their synthesis, we have investigated a multicomponent approach built upon the Catellani platform. Here, we describe a 4-component coupling of aryl iodides lacking an ortho substituent that installs 3 discrete functional groups on the arene in a single step. The process is regio- and chemoselective and uncovers remote substituent effects that have a pronounced influence over inter mediate Pd-(II) complexes. These intermediates have been a long-standing focus of the Catellani-reaction development, but persistent challenges have given rise to the well-known ortho effect. We now show that the ortho effect can be a positive element of reaction design, and that previously problematic iodides can now be used in complexity-generating transformations. In expanding the scope of the Catellani platform, we hope to provide mechanistic considerations to guide future reaction design, while also improving the environmental footprint of synthesizing polyfunctional arenes.

Solvent reorganization is a major driving force of protein-ligand association, but the contribution of binding site waters to ligand affinity is poorly understood. We investigated how altered interactions with a water network can influence ligand binding to a receptor. A series of ligands of the A(2A) adenosine receptor, which either interacted with or displaced an ordered binding site water, were studied experimentally and by molecular dynamics simulations. An analog of the endogenous ligand that was unable to hydrogen bond to the ordered water lost affinity and this activity cliff was captured by molecular dynamics simulations. Two compounds designed to displace the ordered water from the binding site were then synthesized and evaluated experimentally, leading to the discovery of an A(2A) agonist with nanomolar activity. Calculation of the thermodynamic profiles resulting from introducing substituents that interacted with or displaced the ordered water showed that the gain of binding affinity was enthalpy driven. Detailed analysis of the energetics and binding site hydration networks revealed that the enthalpy change was governed by contributions that are commonly neglected in structure-based drug optimization. In particular, simulations suggested that displacement of water from a binding site to the bulk solvent can lead to large energy contributions. Our findings provide insights into the molecular driving forces of protein-ligand binding and strategies for rational drug design.

A method for the palladium/copper-catalyzed direct acylation of azoles with acyl fluorides is described. This study reports the first examples of acyl fluorides being used as acylation reagents in transition-metal-catalyzed aromatic C-H bond functionalization reactions. Depending on the reaction temperature, decarbonylative coupling may also occur.
