The main goal of our study was the synthesis and biological evaluation of 4-amino- and 4-[(4-chlorobenzoyl)amino]-1-methylimidazole N-substituted 5-carboxamides. We investigated the activity of a pyrazolo-isothiazole derivative (G8) against Cryptococcus neoformans. A first screening test showed that G8 at 10 mg/L inhibited the growth of 14 of 15 clinical isolates tested. A series of imidazo[4,5-d]isothiazole nucleosides related to the antibiotic nebularine and the highly cytotoxic 6-methyl-9-beta-D-ribofuranosylpurine have been synthesized from the corresponding heterocycles.
