Enantioselective synthesis, chiroptical properties and absolute configuration of 3-aminosubstituted isothiazole S-oxides

Herein we report a mild and efficient method to synthesize chiral 3-aminosubstituted isothiazole sulfoxides taking advantage of (+)- and (-)-((8,8-dichlorocamphoryl)sulfonyl)oxaziridine under microwave irradiation. The determination of the absolute configuration of the chiral sulfoxide was achieved by theoretical calculation of the CD spectra. A theoretical study of the aromaticity of hypervalent sulfur heterocycles

In this paper we report on the heteroaromaticity of 1,2,5-thiadiazole-1,l-dioxide (I) and its isomers 1,2,4-thiadiazole-1,1-dioxide (II), 1,3,4-thiadiazole-1,I-dioxide (III) and 1,2,3-thiadiazoIe-1,1-dioxide (IV) by calculating their electronic structures and estimating their aromatic character in terms of our previously developed criteria: N, MDQ, Delta E-pi L(NLMO) and Delta E-pi L(Boys) as well as comparing their total energies. (I) is shown to be the most aromatic isomer in good agreement with chemical reasoning and experimental evidence. The 1,l-dioxides of thiazole and isothiazole were predicted to be intermediate in aromaticity between thiophene-l, 1-dioxide and the isomers, (I)-(N). N value measures indicate that thiophene-l,1-dioxide would retain a small measure of aromatic character relative to thiophene (N(thiophene) = 0.868, N(thiophene-l,l-dioxide) = 0.389). A Mild and Efficient Synthesis of 3-Aminosubstituted Isothiazole S-Oxides and their 5-Sulfanylsubstituted Derivatives

The present paper describes a mild and efficient method to synthesize 3-aminosubstituted isothiazole sulfoxides taking advantage of arylsulfonyloxaziridines. The reactivity of the resulting isothiazole sulfoxides toward sulfur nucleophiles has been studied and resulted in the formation of 5-sulfanylsubstituted isothiazoles in a fully diastereoselective way. From 3-benzylamino-5-chloro isothiazole S-oxide an addition-elimination reaction took place affording smoothly the corresponding unsaturated 5-sulfanyl derivatives.

A convenient synthesis of 3H-thieno[3,2-c]-1,2-dithiole-3-thione (7) is proposed. The reaction of 7 with n-butylamine afforded the N-butylthieno[3,2-c]isothiazole-3(2H)-thione (7a) in dynamically equilibrium [1] with its 3H-thieno[3,2-c]-1,2-dithiole-N-butyl-3-imino isomer 7b. Characterizations and antimicrobial activities of the synthesized products are reported.

Under similar conditions, 2-methylbutyl esters of 2-isothiocyanatocarboxylic acids undergo intermolecular oxidative dimerization to give mixtures of thiazolo[5,4-d]thiazoles and 2,3-diisothiocyanatosuccinates. Application of the soft enolization technique to dimethyl alpha,alpha’-diisothiocyanatodicarboxylic esters gives novel cyclic 1,2-diisothiocyanato-1,2-dicarboxylates. Sodium enolates of 2-isothiocyanatocarboxylates, on the other hand, form 5-imino-2-thioxo-1,3-thiazolidine-4-carboxylic esters by nonoxidative dimerization. The mechanisms of the two reaction pathways are discussed.

EDA Complexes of N-halosaccharins with N- and O-donor ligands

A series of EDA complexes of N-iodosaccharin (NISac) and N-bromosaccharin (NBSac) with nitrogen and oxygen electron-pair donors, NISac center dot H2O, NISac center dot THF, NISac center dot Py, NISac(2)center dot Pyz and NBSac(2)center dot Pyz, was prepared and examined by X-ray diffraction and NMR. The halogen-ligand distances are inversely proportional to the donor ability of the ligand. The interactions between ligand and halogen are stronger for iodine than bromine. The X-ray structure analysis has shown that for some compounds the N-X bond in the halosaccharin moiety is not coplanar with the isothiazole ring, and the quantum-chemical calculations demonstrate a high flexibility of the corresponding angle. Complexes were modelled also by DFT calculations using B3LYP and MPW1K functionals. A better fit of the computed geometry was obtained by the geometry optimization in a polar solvent continuum than in vacuum.

The 1,2-dithiolosultam derivative 14 was obtained from the (alpha-bromoalkylidene)propenesultam derivative 9 (Scheme 1). Thus the reduction of the dithiolosultam derivative 14 yielded the alkylidenethiotetronic acid derivative 33 (tetronic acid = furan-2,4(3H,4H)-dione), and the lactam-sultam derivative 10 gave the alkylidenctetramic acid derivative 35 (tetramic acid= 1,5-dihvdro-4-hydroxy-2H-pyrrol-2-one) (Scheme 3). Some of the new compounds (14, 22, 26, and 30) exhibited antimycobacterial activity.
